ERECTA is a kind of stress resistant gene of plants, encoding regulatory proteins that participate in transduction of stress signals and regulate of the gene expression of downstream genes. The gene is involved in the process of growth and development,formation of organ morphogenesis, transpiration efficiency, hormones and light regulation, transduction of stress response signals in Arabidopsis. In this study,ERECTA genes of drought-resistant sorghum were isolated and cloned, and its sequence structure and its expression pattern of response to drought stress at seedling stage were analyzed, respectively. The main results are as follows:(1)SbER1 and SbER2 gene of varieties "Jinza 12" were isolated and cloned, and plant expression vectors φ174-Ubi-SbER1-Tnos and φ174-Ubi-SbER2-Tnos were constructed.(2) The bioinformatics analysis results showed that, the typical protein coded by SbER1 and SbER2 consists of N-terminal signal peptide region, LRR domain,transmembrane region and Kinase domains, and the sequences of LRR domain and Kinase domain are highly conserved.(3) Through the analysis of relative expression gene level of different sorghum varieties by real-time quantitative PCR, the results showed that genes of both SbER1 and SbER2 are expressed in sorghum’s stems and leaves, and the expression level increases with the gradually deepening drought stress. There is alternative splicing of SbER2 genes in different varieties.According to the results of the study, SbER1 and SbER2 can be applied in the engineering in improving the drought-tolerant characteristics of crops. |
