Rapid Membrane Emulsification Technology Application In Veterinary Vaccine Adjuvants

Vaccine is effective to prevent diseases. Adjuvant plays an important role in vaccine, which strengthen antigen presentation, offer deposit effect. Appropriate adjuvant can improve the immunological effect of vaccine. In recent years, research on immunological effect and safety of veterinary vaccine adjuvant has been paid more and more attention. Adjuvant system influence much on immunological effect and safety of the veterinary vaccines.There are two parts of this study.1. Polymer microspheres used as porcine circovirus vaccine adjuvants. PLA-DDAB microspheres with partical size of 1μm and 300 nm were prepared by rapid membrane emulsification. They had small size, good uniformity, which could be adsorbed with porcine circovirus antigen. The microspheres vaccines were inoculated to mice. IgG antibody were increasing duing 35 days after immunization, which significantly higher than that of commercial adjuvants. It maintained a higher level of 210 antibody titers at the 35th day. The 300 nm microsphere also showed advantage in immunology.2. chicken oil emulsion vaccine adjuvants were prepared by rapid membrane emulsification and mechanical stirring respectively. Then we investigated their long-term immunological effect and safety. The membrane emulsification vaccine had higher uniformity. Qualitative and quantitative investigation showed that inflammation caused by rapid membrane emulsification vaccine was lighter and disappeared earlier during 140 days after immunization, which seemed more moderate. Long-term antibody levels showed that membrane emulsification vaccine provides good immune protection to guarantee the immunological effect of vaccine, with NDV and AIV HI antibody titers higher than 26 and 28, respectively.
