Solanum nigrum L. was annual herb and belonged to Solanum, which was a traditional Chinese herbal medicine, the whole plant could be used as medicine, the alkaloids from Solanum nigrum L. could induce apoptosis and inhibit metastasis of tumor cells, the polysaccharides from Solanum nigrum L. could enhance the body’s immunity. Solanum nigrum L. had many clinical roles such as anti-inflammatory, anti-shock, antibacterial and antiviral, anti-inflammatory and expectorant cough and so on.Solanum nigrum L.grew in Heilongjiang Province was used as the experimental materials, the hairy roots were induced by Agrobacterium tumsfeciens A4, C58C1 and A1476, the effects of the factors such as the kind of Agtobacterium tumsfeciens, the explants type, bacterial concentration, infection time, pre-culture time and co-culture time on the induction for hairy roots of Solanum nigrum L.were studied. The bacteriostasis of hairy roots of Solanum nigrum L. on Escherichia coli, Staphylococcus aureus and Bacillus subtilis was studied too, the differences of antibacterial activity on Escherichia coli, Staphylococcus aureus and Bacillus between hairy roots and roots were analyzed.1. The leaf of solanum nigrum L. infected by Agrobacterium tumsfeciens A4, C58C1 and A1476 could induce hairy roots, there were high induction rate for C58C1 and A4,56% and 54% respectively, but the hairy roots induced by A4 grew thick and fast.2. The young leaves, young stems and petioles of Solanum nigrum L. were infected by A4.The results showed that: the induction rate of leaves> induction rate of stems > induction rate of petiole, the induction rate of leaf was 51.7%, the root induced by leaves was also the earliest.3. The leaves of Solanum nigrum L. were infected by different bacterial concentration of A4, when the value of OD600 was 0.6 the induction rate was 58%, which was the highest induction rate.4. The hairy roots were induced by A4 used Solanum nigrum L. leaves at different infection time, pre-culture time and co-culture time. The most appropriate infection time was about 5 minutes.The most suitable pre-culture time and co-culture time were both 2 days.5. The results of PCR detection of Solanum nigrum hairy roots showed, the T-DNA for Ri plasmid of Agrobacterium rhizogenes had been integrated into the genome of Solanum nigrum L. hairy roots6. The hairy roots of Solanum nigrum water extraction had strong antibacterial effect on the 3 kinds of tested bacteria;the bacteriostasis of Solanum nigrum L.hairy roots was significantly higher than that of the real roots on Staphylococcus aureus and Escherichia coli in the 3 kinds of tested bacteria;the hairy roots of Solanum nigrum water extraction had strong antibacterial effect on Staphylococcus aureus and Escherichia coli in the 3 kinds of tested bacteria.The induction system of Solanum nigrum L hairy roots was initially established by this study, and the bacteriostatic experiment was carried out by induced hairy roots, the experimental basis for mass production of Solanum nigrum L hairy roots and subsequent research for hairy roots was provide.
