To discuss wild Oratosquilla oratoria genomic structure and DNA content,Oratosquilla oratoria blood, muscle, liver andhepatopancreas, ovary and testis organizations is materials, and using organizations to analyze Mantis Shrimp the chromosome number, karyotype.Nuclear DNA content and cell cycle of different organizations have been studied by flow cytometry.1. Chromosome number and karyotype analyze of Oratosquilla oratoriaTo understand the karyotype, DNA content and enrich genetics content, this paper analyzed the pattern of karyotype in Oratosquilla oratoria using colchicine injection method for obtaining hypotoniclly fixed testicle, ovary and hepatopancreas specimen. It showed that there were 88 chromosomes in diploid with 2n=88, and the formula of the karyotype could be expressed by 2n=62m+12sm+14t. The total number of chromosome arms(NF) was 162. The maximum relative length of chromosomes is 3.999 and the minimum relative length of chromosomes is 1.623.Do not found that chromosomes are relative to sex.2. DNA content of Oratosquilla oratoriaIn addition, to know the DNA content in different tissues, flow cytometer analyzing was conducted by using DNA of the chicken blood cell as standard. There are two conspicuous peaksin DNA content histogram of testis tissue. One is the peak ofhaploid and another isthe peakof diploid.It is probable owning that there are a large number of sperm and spermatid and a small amount of secondary spermatocyte and spermatogonium.It showed that DNA content was significantly different in different Oratosquilla oratoria issues(P<0.05) and the order of DNA content quantity was muscle>testis> ovary > hepatopancreas > blood. The highest content in muscle reached to 10.43 pg/2c and the lowest content in blood was 8.70 pg/2c. The average of DNA content was 9.61 pg/2c. Moreover, the average value of Oratosquilla oratoria genome size could be estimated as 9400.19 Mbp while 1 pg=978 Mbp. The study is a first step towards broodstock conservation and completely aquaculture of Oratosquilla oratoria in the future.3.Cell cycle of different tissue in Oratosquilla oratoriaBesides, the cell cycle in different tissues is detected with flow cytometer, such as blood, muscle, hepatopancreas,ovary and testis.It showed thathaemocytes in blood has phenomenon of cell cycle like muscle,hepatopancreas andovary. Cell cycleof mitosis in Testis tissue cells is not obvious, but cell cyclemeiosis isvery obvious.Cell cycle of mitosis is the main object inthe study.By flow cytometric analysis,the proportion ofhepatopancreas and ovary cells is lower, however cell proliferating index ishigherin G0/G1 phase. The former is 17.69% and the latter is 12.14%.Cell proliferating index of muscle is low to 5.88%, but thehaemocytes is 7.29%.In this study, using Giemsa analyze Mantis Shrimpchromosome karyotype and take measure of DNA contentin five differentorganizations that is blood, muscle, liver andhepatopancreas, ovary and testis organizationsof Mantis Shrimp.At the same time the study assess its genome size, in order to richthe theoretical basis for Mantis Shrimpgenetics.
