Paeonia ostii‘Feng dan’originated in Tong ling city of An hui province in phoenix mountain area, is a rare treasure in the medicinal peony, for first four generic versions. Paeonia ostii‘Feng dan’peony fung mainly adopts seed breeding, seed cycle for up to six years, seeds and has a very long period, planting to seedling unearthed, generally need to 8 months, the bulls up to 20 months, seriously limits of the traditional Chinese medicine plant expanding production. Key stage in the life cycle, therefore, the understanding of the behavior of seed germination, is a Paeonia ostii‘Feng dan’ important content in the study of biological characteristics. Based on the Paeonia ostii‘Feng dan’ seeds as the material, through the skin permeability, seed extract from different parts of the bioassay, endogenous inhibitory substances bioassay, seed endogenous inhibiting substance and several kinds of separation and identification of monomer inhibition activity test research, obtained the following conclusions:1. The Paeonia ostii‘Feng dan’seeds are deep dormancy type. Mature Paeonia ostii‘Feng dan’embryo structure integrity; Anatomical observations show that kind of embryo structure is complete, but kind of embryo small volume, multiple embedding in the endosperm; Soaking seed vigor is stronger, 54 h after the complete seed and Pierce skin all can achieve water saturated state; Seed extract significantly inhibit cabbage seed germination. Paeonia ostii‘Feng dan’seed dormancy and germination physiological after ripening and contains material, has nothing to do with the kind of skin disorders.2. Using ethyl ether、 petroleum ether、 ethyl acetate、methanol and water as extraction agent to get the seed coat, endosperm and embryo of corresponding leaching, for cabbage seed germination rate and seedling growth, etc. All have different degrees of inhibition. Inhibition from big to small for: root length > seedling height and germination rate. The inhibition degree of leaching solution at different positions of the seed endosperm > seed > embryo. Different leaching leaching agent content of inhibition of ether extraction, the largest petroleum ether is minimal; Ethyl acetate, methanol and water leaching degree by different extraction and determination of the project is not the same. Among them, different leaching agent in the methanol extract 32 kinds of up to 10 organic compounds, Category and the number of organic compounds accounted for 83.33% and 74.42%.3. The system solvent separation method, using ethyl ether, ethyl acetate, methanol and petroleum ether as extraction agent to get the seed coat, endosperm and embryo of corresponding leaching, bullishness of spectrum mass spectrometry analysis(GC-MS), qualitative quantitative detection to 43 kinds of organic compounds, according to the chemical structure can be divided into 12 classes. The number of organic compounds by ester of up to 11, accounting for 25.81% of the total organic acids, ketones and 8 each accounted for 18.60%. Monomer the relative contents of organic compounds in the top 23.19% of organic acids(kind of embryo methanol phase) to 95.19%(embryo) methanol phase, followed by ketones and aldehydes.4. Using the simulation method, according to the seed extracts and relatively higher content of organic compound distribution, select the 1,3- two hydroxy acetone, acetone, phenol hydroxyl, furfural, acetic acid and formic acid in 6 kinds of standard organic compounds, the results of the study show that inhibition activity, the results of the study show that inhibition activity, these organic compounds at the concentration of 1 ~ 10 μl/L, for Chinese cabbage seed germination has significant inhibitory effect, order is from weak to strong inhibition effect: hydroxy acetone, phenol, 1, 3- dihydroxyacetone, furfural, acetic acid and formic acid were gradually increased with the increase of concentration of inhibitor. Longevity long influence to root and sprout inhibition performance is different, 1, 3- dihydroxyacetone(1 ~ 10μl/L), hydroxyl furfural and acetone(1μl/L) processing at low concentration, high concentration treatment showed significant inhibitory effect; Other organic compounds in the 1μl/L treatment significantly inhibited root growth, but relatively high concentrations of treatment between the difference is not obvious.
