People demands more requirements of beef quality on the market because impro vment of living standard and it comes out more and more good quality or high grade b eef products in real life,so developer is requested intense reserch the qulity of beef. A s usual,what we say the quality of meat include meat of nutrition, palatability, and vis ual aspects of content.But in the past,the meat appearance and flavor is not only the m ost concerned by customers but also is the evaluation by these index.With the develop ment of bioinformatics,the animal breeding and genetics and animal reproduction hav e conventional to molecular breeding,meanwhile " gene expression difference" is bein g the reserch hot spot in the genetic improvment and identification of gene function.Using suppression subtractive hybridization(SSH) to build the longissimus dorsi muscle differentially expressed subtractive cDNA library of Chinese red steppe muscle and Chinese holstein cows. 859 differentially expressed clones were achieved. Analysis of the whole library sequencing and comparative with NCBI blasting procedure. The blasting results revealed 789 high quality ESTs, 84 hypothetical function protein gene and 356 functional protein gene. Among functional protein gene, 123 were up-regulated and 233 were down-regulated in hybrids. Positive clones of screening function classification and functional annotation. According to the results of screening the differentially gene function main position in cell, molecular and biological process of these three categories.The clone in the SSH library, we using gene chip technology to validate clone the existence of false positive genetic variations and further screening, screening differentially genes by Sam method, a total of 1296 genes. Ratio=2/5, selected 32 differentially expressed genes, including 24 gene expression were up-regulated and 8 gene expression were down-regulated; Ratio=4/5, screened 43 differentially expressed genes, of which 18 were up-regulated and 25 were down-regulated. Compare with the SSH library, genes differentially expressed the results are consistent. It proved the SSH library has high quality and low rate of false positives.With reference to the national standard of the red steppe and ordinary hybrid cattle fatty acids were determined, the former fatty acid content is higher than the latter. Pick out the differentially expressed genes GLTPD1 and PPARĪ³ which related to function fat metabolism in the library. Using fluorescence quantitative PCR determination of two genes expression in red steppe and ordinary hybrid bull, both in the red steppe expression quantity is higher than ordinary crossbred beef cattle. The correlation analysis in fatty acid and fluorescence quantitative PCR determination results. It showed that GLTPD1 and PPAR gamma can be used as candidate genes of meat quality trait.
