Beta vulgaris is the most important sugar crops in the world, drought stress is an important factor restricting Beta vulgaris production, and cultivation drought-tole ra nt sugar beets can promote the development of the Beta vulgaris industry. Analysis drou ght resistance gene function and Beta vulgaris drought mechanisms is of great significa nce for Beta vulgaris breeding. With the Beta vulgaris genome sequencing’s accomplishment, a large amount of sequence information needs to be identified. Over the last decade, VIGS has been successfully used as reverse genetics technique for gene function analysis in various model plants, as well as crop plants. It has many advantages of analysis resistance-related genes, such as faster, low-cost, high-throughput and it does not require genetic transformation. In order to develop a virus-induced gene silencing system for rapid identification of drought resistance gene in Beta vulgaris, we made the research as follows:1, Through a series of physiological and biochemical indexes of 13 Beta vulgaris germ plasm resources dealing with drought stress, strong and weak drought-resis ta nt material was identified,used for virus-induced gene silencing system establishment and drought resistance gene identification.2, Use TRV to construct beet PDS gene silencing vector,successfully silencing the PDS gene, preventing chlorophyll synthesis in leaves of Beta vulgaris, the leaves were bleached, which is the first application of virus-induced gene silencing in Beta vulgaris.3, Through q RT-PCR analysis of Beta vulgaris Badh, P5 cs, Bip gene expression after the drought stress, P5 cs has strong correlation with drought stress.4, Constructed Beta vulgaris P5 cs gene silencing vectors, Beta vulgaris drought resistance was significantly decreased after P5 cs gene silenced, it proved that virusinduced gene silencing system mediated by tobacco rattle virus can be used to analysis stress resistance genes for functional research.
