Cotton, eggplant, sunflowers, piemarker, mung beans, elm and cabbage etc. seven kinds of agricultural and forestry plants Verticillium wilt samples had been collected from Ili Kazak Autonomous Prefecture Tekes County of Xinjiang, Shihezi grapes station of 121 th farm regiment, and 133 th farm regiment, and 142 th farm regiment and experiment station of agricultural college of Shihezi university in 2013-2014. After the separation and purification were obtained 9 strains of Verticillium wilt, by the morphological and molecular biology(r DNA-ITS sequence analysis), the results show that the 9 Verticillium wilt strains were Verticillium dahliae.Pathogenicity of the isolations were tested through the paper pots tear bottom dipping root methods. Tests showed that the inoculation of crops have the same symptoms as the field disease plants, and uninoculated control plants is normal.The same pathogen as inoculation strain were isolated by re-isolated from the seven kinds of inoculated agricultural and forestry plants.Verticillium wilt strains separated form seven kinds agricultural and forestry plants all can grow on PDA medium during 10 ℃ to 30 ℃, the fastest grow at 25℃, no grow at 35 ℃. Use the microscope observation of each strain conidia size have some difference at different temperatures, but the largest at 30 ℃, conidiophores and microsclerotia of each strain were observed and measured found among conidiophores and microsclerotium of each strain morphology and size were not different significantly. The identified seven kinds of Verticillium wilt caused by Verticillium dahliae, besides cotton Verticillium wilt, Verticillium wilt of eggplant has occur widely in our country, and has been reported early; Verticillium wilt of sunflower has been reported in Xinjiang, Verticillium wilt of cabbage has been reported in Beijing, and Verticillium wilt of other three kinds of crops, namely Verticillium wilt in mung bean, Verticillium wilt in elm and Verticillium wilt in piemarker not reported in China.Nine Verticillium wilt strains from different plant were inoculated on seven kinds of plants, initially, they belong to Verticillium dahliae, but their were difference obviously on pathogenicity. According to the preliminary results of pathogenic test, the nine strains could be divided into the following three kinds of case. First, the strains of cotton, sunflower, piemarker, mung bean, elm, cabbage from the cotton region of Shihezi to cotton, safflower, mung bean are highly pathogenicity and other crops pathogenicity weak. Second, most of test strains to safflower pathogenicity strong, but two strains from eggplant and one strain from sunflower are not infect safflower; Third, Verticillium wilt strains of the same kinds of crops from different ecological zones there are obvious differences in pathogenicity, such as sunflower strain(VD3-1) from the cotton region of Shihezi highly infection cotton and safflower, disease index high up 92.1 and 62.5 respectively, sunflower strains(VD3-2) of Ili Tekes from not cotton region no infection safflower, disease index of cotton is only 6.9. Verticillium dahliae host range is very wide, preliminary study, indicating that Verticillium dahliae might have some special parasitic resistance.Cotton Verticillium wilt is more serious in Xinjiang. Safflower, mung bean, sunflower, piemarker, eggplant, elm and cabbage etc. plants are not recommended when making cotton Verticillium wilt rotation.
