The cultivation of Brassica rapa in our country is very extensive,Brassica rapa is one of the largest consumption in China and has wide cultivation area. Brassica rapa is favored by people because of its rich nutrition, short growing period is and stronger adaptability. While it is important to introduce new desirable traits into Brassica rapa plants using modern biotechnological technique for its short plant, plant small, short vegetative period and low production yield. miR156, which is highly conservative in some plant species, is an important endogenous factor regulating plant growth and vegetative development.This report selects miR156 precursor sequence of cabbage by analyzing cladogram, cloned and constructed fragment that contains mi R156 a stem-loop structure to construct over-expression carrier from the Youqing 49; moreover, it utilizes simulative sequence to establish mimicry156 vectors that swift to Youqing 49 to obtain transgenic plant. In addition, this paper researches the N element content by hydroponic wild-type Youqing 49 whether has the influence to the growth of Brassica rapa,and explore expression of miR156 and target gene at different nitrogen levels to analyze the molecular mechanism.The research results are as follows:1, Analysis of phylogeny illustrated that cabbage miR156 a precursor sequence and arabidopsis thaliana play a major regulation role in ath-miR156 a and ath-miR156 c has the highest affinity;2, Analysis the expression of miR156 in root and leaf of Brassica rapa at different N content treatments through fluorescence quantitative techniques, and discover that there is a significant difference among these expressions;3, Cloned Brassica rapa miR156 a super-expression carrier and target gene simulation mimicry156 expression vector, which successfully transformed Brassica rapa‘Youqing 49’, and obtained the transgenic plants.
