As one of the major crops in China, the high yield and fine quality of soybean has been received much more concern all the time. Also, seeds vigor as its premise safeguard has caused a lot of attention from the experts. In order to explore the essential reasons of different vigor soybean seeds under different cultivation pattern. The spring soybean "chuandou 11(A1)" and summer soybean "nandou 12 (A2)" are used in this experiment. We select two most obvious variation seeds as experiment materials from Al which were sowed at different time and A2 which were took with intercropping program. Then, We investigated the difference of proteome, the content of soluble protein, the vitality of lipoxidase and the glutathione reductase within seeds and sprout during the seeds germination(0h、48h、60h). Finally, we analyzed the relation between these variations and seeds vitality, explored the essence of seeds vitality and built the theoretical foundation for soybeans breeding, harvest and store. The main results are following:1. By two-dimensional electrophoresis, we came out 29 results from 32 variations, examination including 12 protein points restored nutrition,14 enzyme protein points and 2 ion channels of biomembrane. Through reviewing the related articles, we conclude that the enzyme protein point related to seeds vigor is urease, phosphoenolpyruvate carboxylase, glyceraldehyde-3-phosphate dehydrogenase and lipoxygenase. The first three proteins and have positively correlated relationship with the activity of seeds. Urease can increase the seed of nitrogen utilization; while enol phosphate type pyruvic acid can adjust PH value, keep the balance of ion and provide carbon skeleton leguminous plant symbiotic nitrogen fixation. Glyceraldehyde 3 phosphate dehydrogenase take part in the pentose phosphate pathway and response to oxidative stress, etc.2. By comepare with the different vitality’s Protein point in a varieties:During the dry seed period, spring soybean, the sichuan bean boost prices phosphoenolpyruvate carboxylase, and two urea enzyme addition. Low vigor seed have lipoxygenase differences; while in summer soybean, the south beans 12 net have four glvceraldehyde -3-phosphate dehydrogenase variations among the different cantons. At 48 h imbibition, the main difference between chuan bean 11 and south bean 12 is that, the former soybean contains globulin point, while lipoxygenase from interplanting appear in the later one. For the 60h imbibition, high vigor seeds contain new synthesized soybean globulin point,,while south bean 12 have glyceraldehyde 3-phosphate dehydrogenase.3. The correlative character between the soluble protein content of soybean and the seeds vitality would embody clearly only on the condition that the soluble protein content reaches a certain critical value. This outcome conformed with the results of "There is no correlation between the amount of solute protein and the seeds vitality" by Zhang Wenming, and also correspond with the results of "The seeds vitality has a positive correlation with the amount of solute protein". During the soybeans germination, There is a positive relation between the seeds vitality and the speed of soluble protein degrading, indicating that the higher vigor the seeds show, the faster they synthesize during the germination progress.4. The activity of lipoxidase is lower in high vigor seeds than low vigor seeds, as well as in summer soybean seeds and spring soybean seeds. But the activity of lipoxidase tend to increase during germination, the increasing activity of lipoxidase is faster in summer soybean than spring soybean. The free radical production conducted by reaction of lipoxidase can destroy the membrane lipids of dried seeds leading to the acceleration of seeds deterioration and reduction of seeds vitality. However, the transportation of new substance within cells would benefit from the damage of membrane lipids.5. In the period of bud, the activity of glutathione reductase show the variety within and between the species of spring soybean and summer soybean, the glutathione reductase in high vigor seeds is more active than that in low vigor seeds within species. During the germination, the effect of glutathione reductase in high vigor seeds raised, while the low vigor seeds act in an exactly the opposite way. As an important element in enzyme protected system at dried seeds period, glutathione reductase can eliminate reactive oxygen species effectively in case of seeds deterioration.
