| Since the Equus Przewalskii introduced into our country, Gasterophilus has become a important biological factor that limit the horse wild-release successful. That is very important to understand the biological processes of Gasterophilus in vitro development of its host. The study aimed at the scientific problems of Gasterophilus in Xinjiang Kalamaili nature protection area, such as many unknown of the biological processes of Gasterophilus in vitro development of its host and the threshold temperature and effective accumulative temperature. Research using the field investigation combined with laboratory experiments, and the conclusions are summarized as follows:(1) The field investigation for local horse in conservation area has been done from March to April in 2013, checking 173 fresh faeces in 23 days. Respectively on March 19, March 27 and March 31 found one G. nigricornis mature larvae, other Gasterophilus species mature larvae were not found during the investigation. Compared the results with previous survey study, we can suggest that March to April is not the peak discharge period of Gasterophilus mature larvae. Combined with the activity routines of local horses, we speculated the local horses are not the repeated source of infection for Gasterophilus of Przewalski’s horse.(2) The field investigation for survival of natural discharge of Gasterophilus mature larvae in faeces of Przewalski’s horse in conservation area from June to July in 2014, we found 16 natural discharge G. pecorum in 211 fresh faeces. The larvae survival rate was 100% which found in the surface of faeces, while the survival rate of larvae in the faeces can only be 29%, there was a significant correlation between the survival rate and the location in the faeces. We speculated that the cause of death of the larvae are more likely to be the position in the faeces.(3) According to the date of pupate,69 mature larvae were divided into 10 groups, cultivated larvae under the natural variability temperature conditions indoor. The developmental threshold temperature and effective accumulated temperature for pupae of G. pecorum were determined by using rule of effective temperature summation "least square method". The results shows that:the developmental threshold temperature had a range of (10.58±0.64)℃ and the effective accumulated temperature had a range of (190.95±24.85)d℃. Unary linear regression equation was established between the developmental temperature and developmental rate:(V=-0.0541+0.00527). The linear regression analysis showed a significant relationship between the developmental temperature and developmental duration(P=0.001). The development duration prediction for pupae of G. pecorum was established:N=(190.95±24.85)/[T-(10.58±0.64)].(4) Using G. pecorum mature larvae from alimentary canal of accidental death Przewalski’s horse, we choose 30 G. pecorum which successful pupated for the feather experiments and set up human disturbance. Result shows that the total eclosion rate of larvae G. pecorum mature larvae from alimentary canal of accidental death Przewalski’s horse was 53%. Correlation analysis was carried out on the experimental results, we studied the impact of human disturbance had no obvious effect on the feather. |
