On the basis of preliminary field survey, surveyed the resources ornamental characters and heat resistance form field investigation of 26 pansy from April to July in 2014, and researched heat-resistant resources physiological and biochemical characteristics under high temperature stress of different pansy.Cloned the HSP70 gene and studied the expression of HSP70 gene under high temperature stress. The main results as follow:(1)The results of phenotypic traits showed that there was a large difference between stem length, plant width, flower diameter, leaf length, leaf width, branch number, flower number, design and color of 26 parts pansy. Combined with high temperatures pansy morphology and survival, 3 parts of screened resources have better heat resistance characteristics, namely HAR, DFM-16, DFM-17.(2) The photochemical efficiency of 5 parts pansy were all fell under high temperature stress.and the heat resistant decline was little than not heat resistant resources.Both the cell membrane permeability and heat pollution index were low, and the proline content rise, showed that a stronger adaptability under the high temperature.The POD and CAT activity were lower than control, while SOD activity remained relatively stable under high temperature stress.(3)Obtained HSP70 gene fragment of 4 pansy resources, the length of E01,DFM-16,DFM-17 were 1571 bp, the 08 H was 1572 bp,the sequence alignment showed that the sequence similarity was 98.33%.The similarity with HSP70 gene of reference and Arabidopsis were 93.06% and 74.13%, and above 84% with castor oil plant,cacao and other plants.The gene were located in conserved regions and similar high rate, they were homologous genes. According to the characteristics of HSP70 gene conservative and base sequence similarity rate, the four genes were pansy HSP70 fragment.(4)There was no difference between heat resistant pansy and not heat-resistant pansy of HSP70 gene expression under normal temperature processing.With the temperature increasing, the higher amount of HSP70 gene expression, and the DFM-16 was 2.5 times higher than the control under 42℃;With the increase of the processing time, the DFM-16 and 08 H gene expression of HSP70 had no significant change under room temperature, with the increase of the processing time, DFM-16 and 08 H were sharp rise in the amount of the expression of HSP70 gene under 42℃,and about 6 and 15 times than the control.The heat resource DFM-16 was significantly higher than 08 H..Showed that HSP70 gene differential expression was one of the reason which caused by different heat resistance difference between pansy resources.
