There is high nutritional value and rich minerals in milk, which includes calcium,phosphorus, iron, zinc, copper, manganese, and molybdenum. The main ingredient is protein and fat in milk, the milk fat and milk protein are paramount nutritional indicators. Importantly, milk is the best source of calcium to human, and milk was known as "white blood”, because it has the appropriate proportion of calcium and phosphate, and is beneficial for the absorption of calcium. With the rapid development of China’s dairy industry, high quality milk was needed by the people, the focus and hotspot of the geneticists’ research is promoting the milk quality. Holstein cow was the main varieties of producing milk, the study of cow milk traits was determined,include milk fat, milk protein, somatic cell count, milk production, lactose, and dry matter. These performance measurement data were obtained from a total of 15 Chinese Holstein cow. Milk fat rate is the focus of the study. 3 high-fat cows and 3low-fat cows were chosen from the 15 Chinese Holstein cows by performing vivo experiment for verifing its related functions. This study laid a better foundation for screening the related genes which associated with milk quality and establishment of molecular breeding method.ACADSB gene is the differentially expressed genes which has been chosen by our previous transcriptome analysis and associated with the milk fat metabolism of cows. It is important of further study to the correlation between ACADSB gene and milk fat metabolism on dairy cows.This study was to build ACADSB gene RNAi vector and over-expression vector,and then transiently transfected into mammary epithelial cells of cows, and verified at the m RNA level and protein level. Meanwhile, the intracellular triglyceride content(TG) was determined. The results showed that over-expression of ACADSB gene could significantly increase the level of intracellular triglyceride, and silented ACADSB gene could significantly inhibit its content by constract. On this basis, 3high fat cows and 3 low fat cows were chosen from 15 healthy cows, which were in the same environment and in the lactation period for this vivo experiments, and then the total RNA was extracted from the mammary tissue, reverse transcribed RNA into c DNA by RT-PCR, and the ACADSB gene expression was detected by q PCR in different mammary tissue of cows. By the effect of ACADSB gene on milk fat metabolism, it laid a study foundation of Chinese Holstein cow’s genetic breeding, it also pointed out the direction to better build a low-fat or non-fat cows group. The study explored systematically the effect of ACADSB gene on the milk fat metabolism,and the expression of ACADSB presents a positive correlation with the content of milk fat, it also may regulate the metabolism of cellular triglyceride.
