As one of the most important coal production bases in China, mine area reclamation have been done in Shanxi Province for many years, but the situation is still serious. The key point is to improve the soil conditions and restore land reclamation. Mining area is seriously lack of potassium which is necessary for plants growth and the potassium bacteria plays an important role to restore the use of soil potassium. This paper screened strains from mining reclamation soil of Xiaoyi,Changzhi and Gujiao, then identified the strains, and made pot and simulation experiment to study the influence of strains to soil nutrients and crop growth. All those experiments were able to screen effective strains for mine area reclamation and rich the bacteria resources.Twenty-six strains were screened,the colony and cell morphology of strains were similar, the characteristic of colony morphology were circular, moist smooth, neat edges,thick colonies.Used hydrogen peroxide ashing method to calculate the releasing rate,five strains were picked out (K15、K17、 K19、K24 and K26),their releasing rate were 21.18%,22.29%,27.89%,23.4%, 17.89%,higher than GSY (mucilaginousus bacillus) obviously,and K19 was the highest one.The five strains were all G,rod-shaped, capsule,and K19 with long rod,spores,the others with relatively short rod, and no spore.By cultivating the five strains of bacteria,it was found that these bacteria could grow well in ashby medium,organic phosphorus or inorganic phosphate synthetic medium.The five strains can use sucrose, glycerin, mannitol and starch as carbon source,use of nitrate and ammonium as nitrogen source,they could also grow in the pH7~11,and 1.0%salt medium.They produced ammonia,indole,their catalase, sugar fermentation were all positive,their Methylred, VP, oxidase, citrate utilization,Phenylalanine deaminase,hydrogen sulfide were all negative.K19 was relatively similar to GSY,they can’t make nitrate reduction,produce lecithinase,and breed best in nitrogen-free medium.By using Biolog method and 16S rDNA sequencing method to identify the five strains,it showed that K15, K17, K24, K26 were Sphingobacteriaceae, K19 was paenibacills mucilaginosus.K19,K24 were used on the mine soil to make pot experiment and simulation experiment, the results showed that the bacteria was conducive to the growth of plants and the accumulation of material quality, increased the plant accumulation of Nitrogen, Phosphorus and Potassium,and also available potassium, slowly available potassium in mining soil.It could also increase the enzyme activity of soil urease, catalase, and invertase and the number of bacteria and actinomycetes in soil,changed soil microbial community diversity.K19 strain made significant function in improving soil fertility and crop quality,it has reached the effect of GSY and potassium chloride fertilizer,and showed more superiority.This research discover that K19 as one of the good potassium bacteria can be used as the effective biological bacterial fertilizer.
