Pear is a typical fruit tree of gametophytic self-incompatibility (GSI). This characteristic results in low yield under the condition of natural pollination, so it is necessary to arrange varieties for pollination or to perform artificial pollination toincrease the yield. This research use pear as materials, Based on specific PCR and sequence clone, self-incompatibility genetype (S-genetype) of some pear cultivars and a few types of wild species native to China were identified.S-RNase specific PCR were used with "FTQQYQ" and "anti-IIWPNV". After cloning and sequencing of PCR products, the S-genotypes of 20 cultivars or wild-species from China were determined:Xiangshui 517S31,Zaoxiangshui S26S42,Shanli No.3 S33S41,5hanli No.5 S4S42. Yunnanmali No.l S9S42, Yunnanmali No.2 S30Sx, Douli S30S31, Mianli S19S41, Red Comis SeSd, Chihuali S26S15, Feb.15th S4S12, Nov.11 S2S26,Saozhoumiaozi S15S26, Anhan No.3 S13S31, Xingchengxiehuatian S17S31, Baibalixiang S19S31, Pitai S22S43, Huangjinduima S19S29。To identify S-genotypes of pear cultivars or a few types of wild species native to China. S-RNase specific PCR were used. After cloning and sequencing of PCR products, the S-genotypes of 18 cultivars or wild species from Gansu were determined. The results are as follows:Linxiaxiangba S12S2、 Ganguheili S16S64、KanglebaiguoSbS、Wudutianli S26S、Linxiahuangma S3Se、Kanglesumuli S1Sh、 Linxiasala S16Se、Jiuquanchangbali S6S22、Linyaomatianli S22Sd、Jiuquanmaili S6S17、Manluwowoguo S12S21、Wushantangli S8S19、Kangleganchangba S22Sd、Hezhengganchangba S22Sd. Ganguxiangshui and Lixianxinbapan has a Se-RNase gene. There is a S26 in Linxiadadiaodan and S16 in Ganguhongxia. Subsequent sequence analysis showed that there is a novel S-RNases in Ganguheili, which is named S54.In order to make sure the S-genotypes of individual species with different reported results, Two pair of primers specific to pear S-RNases, PCR amplification of Sgenes fromgenomic DNA of 37 specimens from different place of fifteen pear cultivars were carried out. Then the PCR products were extracted, cloned and sequenced. The amplified fragments were assigned to their respective S-RNases by Blast analyses. The S-genotypes of the fifteen tested pear cultivars were identified as follows:Baozhu S22S42, Cangxixueli S5S15, Hanxiang S12S31, Hangqing S1S4, Jinqiuli S3S9, Jinfeng S17S19, Longxiang S16S42, Nanguoli S11S17, Pingxiangli S31Sd, Qingkui S1S3, Qingxiang S4S7, Qiubai S17S19,Yaqing S4S17, Yanbiandaxiangshui S12S16, Zaoli 18S4S28...
