| Protein-protein interactions play a pivotal role in coordinating many cellular processes. Recently, exploring protein-protein interactions and the regulatory function has become a research hotspot. Wheat TaJRL2.1 gene, which could be induced by Fusarium-treatment, and participated in wheat scab resistance. In previous studies, by using yeast two-hybrid system, we identified 43 TaJRL2.1 interacting proteins. This study was to verify the interacting proteins and analyse their expressions.Firstly, BiFC technology was employed to further verify the interaction between TaJRL2.1 and the 11 TaJRL2.1 interacting proteins that screened by Y2H in vitro. Among them, five proteins including glycolic acid carboxylase GLO, ribulose bisphosphate carboxylase activase RCA, WRKY transcription factor, protein phosphatase PP2C and pollen-specific protein PLIM-like can be detected to interact with TaJRL2.1 in the onion epidermal cell membrane, nucleus, cytoplasm and some organelles. Secondly, we analysed the function of the interacting genes, found that all the five genes were induced by Fusarium infection. In TaJRL2.1-OX transgenic plants, four interacting genes (GLO, RCA, WRKY, PP2C) were also upregulated significantly in addition to the reduction of PLIM-like. Attenuating WRKY expression through virus-induced gene silencing led to the down-regulation of PAL and C3H which both were involved in ligin biosynthesis. All these results laid the foundation for further revealing the scab resistance mechanism of TaJRL2.1. |
