Cucumber angμLar leaf spot caused by Pseudomonas syringae pv. lachrymans is distributed worldwidely and inflicts different degrees of damage, which can also induce bacterial spot of melon leaves. It was typical seedborne pathogen. Pseudomonas syringae pv. lachrymans has become great threat to cucumber and melon industry, For this reason, ensuring the exclusion of infested seeds by applying the effective, commercially viable and convenient detecting technologies is crucial for the food industry.Bacterial fruit blotch is the most important melon seed-borne diseases, the Pathogen is Acidovorax citrullia, which is a worldwide quarantined disease. melon infected Acidovorax citrulli and Pseudomonas syringae pv. lachrymans,the symptoms very similar to each other. It is difficμLt to use traditional methods distinction between the two.In the present study, Padlock and LAMP assay were developed for detection P. syringae pv. lachrymans strains from cucumber and muskmelon.We detected the specificity, sensitivity seedlots with artificial infestation and applicability of seed samples. To adapt padlock technology for diagnostic purposes, we combined the probe and Macroarray technology. The resμLts showed that:The two probes and two sets of LAMP can both distinguish the two groups strains from all other experimental strains, and seed pathogen carrier rate was 0.1%(1/1000). The probe technology combined with Macroarray was used commercial melon seeds and cucumber seeds detection,7 of 205 melon seedlots collected from Xinjang province were positive for Pseudomonas syringae pv. lachrymans strains from melon,and 19 cucumber seedlots was not detected Pseudomonas syringae pv. lachrymans strains from cucumber. The detection sensitivity of the pathogen was up to100fg/μL for Padlock and LAMP.We designed One specific padlock and one specific LAMP to melon strains, Which can both distinguish between the Acidovorax citrullia and Pseudomonas syringae pv. lachrymans strains from melon. Providing more method to detect Acidovorax citrullia,The two specific probes and two specific sets of LAMP distinguish the two groups strains from all other experimental strains. combining probe with Macroarray established the high-throughput detection of syringae pv. lachrymans. which further confirmed the probability that the two groups strains belonged to different pathvars.
