Cell wall, which is involved in plants water absorption, nutrient transport, and the interaction of plants and environment, is a particuliar constituent of plant cell. It has demonstrated that mutiple gene families involved in cell wall biosynthesis plays an important role in the process of cell wall extension, thickening and modification. Besides, OsTHEl gene family is a receptor-like protein kinase subfamily participating in rice cell-wall synthesis. In this article, the structure and function of OsTHE1 family genes were predicted through bioinformatics analysis, and the functions of OsTHEl family genes were analyzed with qRT-PCR and related gene mutants. The main results are as fellows:1. Bioinformatics analysis of OsTHE1 gene family shows that, it contains 19 members, of which can be divided into 2 subgroups. The protein family contained signal peptide,1-2 carbohydrate binding domains, transmembrane domains and intracellular kinase domains. In addition,17 genes have no introns. In addition, the most genes of this family contain cis-acting elements related to light signal induction expression and endosperm development in their promoters.2. The OsTHEl family genes expression microarray and qRT-PCR analysis showed that, these family genes can be divided into two groups with three subgroups. qRT-PCR analysis confirmed OsTHES6 constitutive expressed and OsTHES11 anther-specifically expressed. In addition,8 gene expression levels were markedly increased in the 3rd internode compared to the 1st and 2nd internodes, implying their involvement in the secondary cell wall biosynthesis.3. T-DNA homozygous mutants of OsTHES2, OsTHES6 and OsTHES16 and T-DNA heterozygous mutant of gene OsTHES11 were obtained by genomic PCR and hygromycin-resistance detections..4. OsTHES6 Over-expressing vector was constructed, and then its positive transgenic plant was generated. Moreover, over-expression vectors with FLAG tag protein gene and RNAi suppression expression vectors of OsTHES10 were also constructed.5. Sensitivity resonse of seedling to light/dark growth experiment showed that, the OsTHES6 overexpression T1 positive transgenic plants and T-DNA homozygous mutants were treated under light or dark, the OsTHES6-overexpression reduced the sensitivity to light treatment while its insertion mutatant increased the light sensitivity. It suggested that, OsTHES6 gene is involved in the process of light response, corresponding to the cis-acting elements in its promoters.In this paper, the evolution, gene expression and functional identifications of this gene family were also discussed.
