In this study, experimental materials areBrown Tsaiya Ducks,FSH/3 is selected as candidate gene. We used PCR-SSCP to detect the association of single nucleotide polymorphism of FSHβ gene andanalyze its polymorphismgenotypes with production traits. qPCR and 2-ΔΔCt method were applied to detect the copy number variation of FSHβ gene and analyze the association of copy number variation of FSHfβgene and their combined genotypes with production traits.Theprimaryresults were as follows:(1) There were 1 mutation was detected in FSHβ gene and three genotypes were identified.H allele frequency was 0.3191 and h allele frequency was 0.6809, the gene loic heterozygosity was 0.4346, polymorphism information content was 0.3401 and its belongs to an intermediate polymorphism.χ2 test suggested that the population was in Hardy-Weinberg equilibrium. Analysis showed that the individuals of genotype Hh and hh were significant lower thanthat ofgenotype HH in weight of four weeks of age (P<0.05); the individuals of genotype Hh and hhHH were significanthigher thanthat of genotype HH in egg weight(P<0.05); the individuals ofgenotype Hh were extremely significant lower than that of genotype HH in shell thickness(P<0.01), while the genotype hh also significant lower than that of genotype HH (P<0.05).(2) Primers of FSHβ and Anpl-DRA were designed and standard curve was established. The standard curve ofFSHβ and Anpl-DRA showed that the correlation coefficient were 0.996 and 0.990, respectively.The amplification efficiency of the standard curve ofFSHβwas 108.337% and the slope of standard curve was-3.137.The amplification efficiency ofthe standard curve ot Anpl-DRA 106.068%and he slope of standard curve was(3) In this research, five kinds of copy number variation were identified and the population number of CNV type is 42、26、15、7 and 4. Association analysis of production traits showed that the individuals with five kinds of copy number were significant correlation in eggshell thickness (P<0.05), the individuals withlVcopy numberwas significant lower than that of ⅠⅡ、and Ⅲ copy number in protein level (p<0.05).
