Through the analysis of maize changes on agronomic traits and yield traits, study the influence of Saccharomyces boulardii on Maize growth, Finally, the Saccharomyces boulardii on maize leaf spot, leaf blight, leaf blight pathogen in vitro antibacterial test, Analysis of the inhibitory effect of Saccharomyces boulardii on three maize diseases, it hopes to play a reference role for the study later fermentation of Saccharomyces boulardii, and application of Saccharomyces boulardii. The main results are as follows:1 The optimization process of Saccharomyces boulardii fermentation:(1) The components and culture conditions of optimal medium YEPD(A),which were obtained by single factor experiment: Glucose 8%; NH4H2PO4 0.7%. PH 5, temperature 30 ℃, inoculation amount of 1%; speed 200r/min, liquid volume 50/250 ml, culture 24 h.(2) The best liquid medium based on YEPD medium from quadratic rotation orthogonal experiment were as follows: Glucose 1%, Yeast Extract 1.5%, Peptone 3%, KH2PO4 0%, Mg SO4 0.1%, 30 ℃, culture 24 h.(3) Orthogonal test L9(33)optimization based on single factor results YEPD(A), the optimum medium YEPD(B) was obtained: Yeast extract 1%, Glucose 8%, NH4H2PO4 0.2%, 30 ℃ culture 24 h.(4) Saccharomyces boulardii nutrient rich solid fermentation fermentation process optimization results: p H = 5, bottling volume at 12 g / 250 m L base material(3.6 g soybean meal, 3.6 g of bran, 4.8 g of corn flour) inoculated amount of 0.5%(600 μL), 15.6 m L of distilled water, 2% glucose, 1% peptone,Mg SO4 0.01%, K2HPO40.05%.(5) Saccharomyces boulardii barren solid state fermentation medium optimization results: The five different kinds of culture medium(respectively: PDA, YEPD(A), YEPD(B), Sabouraud medium, Czapek medium) for Saccharomyces boulardii seed liquid, inoculated into a barren media(solid state fermentation medium barren: maize flour 5 g, bran 100 g, soybean meal 4 g, water 109 ml 121 ℃, 20 min), YEPD(B) the maximum concentration of bacteria obtained, That is to say five kinds of culture medium, YEPD(B) is most suitable for poor solid substrate fermentation of Saccharomyces boulardii seed liquid.2 The application results of Saccharomyces boulardii in Maize Production:(1) Effect of soaking time and concentration combination test on seed vigor:With distilled water and resuspended after centrifugation of Saccharomyces boulardii suspension precipitation maize seeds soaked for 1 h, can improve the Tianjin Agricultural 14-1 seeds of maize seed vigor.(2) The test results of fermentation products of Saccharomyces boulardii in growth in Maize field:a) Influence of agronomic traits: each treatment on the number of emergence day, heading for several days, silking days, plant height, ear height, impact bald, empty bar, diseased plants and other agronomic traits is not significant, soaking + wash seed treatment group relative the control group were significantly different impact on the properties of fold down, pour fold increase in the number;b) Effect on yield Traits: each treatment on bald, ear length, yield, kernel weight, test weight, kernel rate, average fresh ear weight, ear diameter, the impact of differences in grain yield and number of characters is not significant. In the yield traits of various data,, the soaking treatment on yield, although not significant, but in all treatments, seed soaking for all yield traits are maximized.3 Saccharomyces boulardii on corn leaf blight, leaf blight, leaf spot pathogen inhibitory effect: Antibacterial tests showed that Saccharomyces boulardii suspension on three maize pathogens did not produce inhibition zone, ie no inhibitory effect.
