Background and aim:Cadmium pollution is a serious threat to human,animal and plant health.Cadmium (Cd) chronic poisoning has become one of the important reasons for the disturbance of livestock reproduction.The main reason that induces the apoptosis of testicular cells is the production of reactive oxygen species (ROS) and lipid peroxidation damage of cell membrane.Signal Nrf2 antioxidant response can be induced by antioxidant compounds sulforaphane (SFN).This experiment was conducted to study of sulforaphane by protective of cadmium induced mouse testis support cell can be achieved by Nrf2-ARE pathway.It is providing a new idea for the treatment of oxidative damage induced by cadmium in germ cells and also providing theoretical reference for the prevention and treatment of cadmium poisoning.MethodrTreatment of mouse testis cells with different concentrations of SFN and CdC12 alone or in combination.MTT method was used to detect the survival rate;The content of MDA and the activity of SOD and LDH were detected by colorimetric method;Apoptosis rate was detected by AO/EB double staining and flow cytometry;qPCR and Western blot were used to detect the expression of Nrf2, HO-1, GSH-PX and its regulation of y-GCS, NQO1 gene mRNA and protein.Result:1.CdCl2 can reduce the survival rate of cells and low concentration of SFN can increase the survival rate of cells.2.CdCl2 increases the LDH activity of cells (p<0.01),decreases the activity of SOD(p<0.05), increases the content of MDA (p<0.05);SFN can reduce the LDH activity of CdCl2 cells (p<0.01),increases the activity of SOD(p<0.01,decreases the the content of MDA(p<0.01).3.CdCl2 induces apoptotic rate of sertolis cells increase significantly(p<0.01),SFN can inhibit the apoptosis of cells induced by CdCl2(P<0.01).4.CdCl2 reduced the expression of Nrf2 downstream target genes and and protein (P<0.0l, p<0.05),SFN increased the expression of Nrf2 downstream target genes and protein with treated by CdCl2 (p<0.01, p<0.05).Conclusion:When cells were exposed to cadmium and the sulforaphane has a protective effect on the injury and apoptosis,it can activate the Nrf2-ARE path and up regulation of the expression levels of its downstream antioxidant and detoxification enzymes to inhibit cell damage caused by cadmium.
