Haemophilus Parasuis Typing And HigBA Prokaryotic Expression

HPS often colone on porcine respiratory tract, in the specific conditions, which often causes diease.The disease is called Glaser’s disease. HPS is a gram negative bacillus pumilus, changeful form, which is divided into 15 serotypes, needing NAD, no hemolysis phenomenon, around the Staphylococcus aureus growing well, showing a satellite phenomenon. HPS each serotype virulence is obviously different, of which the most virulent strains for serum type 1, 5,10,12,13,14 type, moderately virulent strains of type 2,4,8 and 15 type, type 3,6,7,9, 11 is a weak strain and did not cause clinical infections. Studies have indicated that the TA system of the Haemophilus influenzae and the Acinetobacter Bauman is helpful for its retention in the host body.Hps isolated from Beijing surrounding does serological typing and prokaryotic expression, protein purification, functional analysis and immune primary analysis of Haemophilus parasuis HigBA gene. Through the construction of pET-30a (+) prokaryotic expression vector, the prokaryotic expression system is used to express the HigB and HigA, and the protein is purified by the combination of the His tag and the nickel column. By growth inhibition test, HigB and HigA are proved to form TA system. The expression of HigB was increased under high temperature and high density pressure environment by qRT-PCR. HigA and HigB protein immunization of guinea pigs, guinea pigs obtain 30% protective force.
