The Detection And Research Differentiation Isolates Of Cucumber Mosaic Virus

Cucumber mosaic virus (CMV),the type species of the genus Cucumber in the family Bromoviridae,is the most widespread plant virus causing disease in various crop and non-crop plants in China and worldwide.Investigate the CMV’s spread trend, and analyse phylogenetic relationship among different CMV isolates had been considered in this study.The main results are as follows:1.In 2013-2014,1367samples in Solanaceae,Cucurbitaceae,Cruciferae and Leguminosae were collected from Hunan,Qinghai,Xinjiang and Hainan province. The samples were identified by RT-PCR and ELISA.The results showed that the positive rates of CMV were 35.70%, positive rates of CTV were 20.33%, and complex infection phomenon was exsited in many samples,which were 15.61%.2.CP,MP and 2b gene of different CMV isolates were cloned respectively. Compared the three genes with sequence on NCBI,their identity were 96.13-99.93%, 97.34%-98.97% and 95.21-98.11%,respectivity.3.15CMV isolates were selected on analyze their phlogenetic relationship and RT-PCR-RFLP research of CP,MP and 2b gene,respectively.The results showed that 13 isolates belong to Subgroup IB,1 isolate CMV-Q5 belong to Subgroup II while 1 isolate that its CP and 2b gene belong to subgroup IB,the CP and 2b gene of CMV-N7 have 97.74% and 98.09% identity with other isolates,but MP gene belong to subgroup II and MP gene of CMV-N7 has 80.04% identity with other isolates.We found the gene of CMV-N7and CMV-Q5 are of the same model by using RT-PCR-RFLP analysis;the biological characteristics of CMV-N7 and CMV-Q5 were significantly different from CMV-P3613 on the system and necrosis host.And electron microscope showed that the arrangement system of virus of CMV-N7 and CMV-P3613 were different.We speculated that CMV-N7 may exists rearrangement or recombination between the group IB and II which was infected Pathogenic ability.
