Cnaphalocrocis medinalis(Lepidoptera, Pyralidae) is one of the most important migratory pests on rice and other crops. It’s frequently sudden outbreak seriously affected the production of rice. In addition to environmental factors, endocrine regulation factors such as juvenile hormone regulation is one of important factors in the regulation of insect migration and reproduction. In all the insects,the JH titer is regulated mainly by biosynthesis pathway and degradation pathway. In this paper, we study on juvenile hormone synthesis pathway related genes of the rice leaf roller, It is helpful to further reveal the mechanism of the migration process.1. Cloning and analysis juvenile hormone synthesis related genes in Cnaphalocrocis medinalisThe CmAACT sequence length is 1332 bp, encoding 330 amino acids, which contains the ”thiolase” domain; CmHMGS sequence length is 2046 bp, encoding 456 amino acids, which contains two conserved domain “HMG-Co A-S-euk” and “init-cond-enzymes”; CmHMGR sequence length is 2710 bp, encoding 654 amino acids, which contains two conserved domain “HMG-Co A_reductase_classI” and “2A060605”; CmMK sequence length is 1992 bp, encoding 228 amino acids, which contains a conserved domain “GHMP kinasesN”; CmMPK sequence length is 1243 bp, encoding 193 amino acids, which contains a conserved domain “P-mevalo_kinase”; CmMPPD sequence length is 1399 bp, encoding 389 amino acids, which contains a conserved domain “mevDPdecarb”. In this study, we found two CmFPPS, CmFPPS1 and CmFPPS2, both contain conserved domains of “Trans-IPPS-HT”, CmFPPS1 sequence length is 1782 bp, encoding 428 amino acids, CmFPPS2 sequence length is 1327 bp, encoding 360 amino acids. To construct the phylogenetic tree, CmFPPS1 and Cm FPPS2 belong to two different clades. CmJHAMT sequence length is 1418 bp, encoding 272 amino acids, which contains two conserved domain “AdoMet-MTases” and “Methyltransf-31”.2. Expression analysis of juvenile hormone synthesis related genes in Cnaphalocrocis medinalisThe results of real-time fluorescence quantitative PCR showed that CmAACT, CmHMGS, CmHMGR, CmMK, CmMPK, CmMPPD, CmFPPS1, CmFPPS2 and CmJHAMT were both expressed in the pupa and adult stage, 3 days old emerged females have a peak of expression these genes. In addition, CmHMGS, CmHMGR, CmMK, CmFPPS1, CmJHAMT in emerged females on day 7 also has a peak of expression. In this paper, we found juvenile hormone synthesis pathway related genes of the Cnaphalocrocis medinalis were expressed in head, thorax and abdomen tissues, and three of the gene expression in the abdomen is very high, such as CmHMGS, CmFPPS1 and CmJHAMT.The qPCR results from comparing juvenile hormone synthesis pathway related genes expression in normal and low temperature with short photoperiod populations of Cnaphalocrocis medinalis show that CmAACT, CmHMGS, CmHMGR, CmMK, CmMPK, CmMPPD, CmFPPS1, CmFPPS2, CmJHAMT have different expression frame,and low temperature and short photoperiod effect these genes expression.
