Antioxidant is an important physiological mechanism to maintain the nomal metabolism and healthy defense. We all know that, different antioxidants have different molecular structure, and their antioxidant mechanism is not the same. Many researchers used to determine the activity of single antioxidant in different system. We can not compare the activity of different antioxidant, because the test system can affect the antioxidant activity.This study was conducted to compare the activity of 5 kinds of antioxidant in the same system.Experiment 1: Comparetive study of diffierent kinds of antioxidant in vitroEvaluated the antioxidant activity of tert-butyl hydroquinone(TBHQ), tea polyphenol(TP), vitamin E(VE), pyrroloquinoline quinone(PQQ), grape procyanidine(GPC) in vitro by determining the total antioxidant activity, the ability of scavenging DPPH free radical and the antioxidant activities of the antioxidants in the rapeseed oil and soybean oil stored at different temperature 20 °C and 40 °C, respectively. The results showed as follws: 1)this five kinds of antioxidant had the same results in antioxidant activity and scavenging DPPH, and they were TP>GPC>TBHQ>VE, PQQ could not react. 2)the effect of five kinds of antioxidant on the rapeseed oil and soybean oil stored at different temperature 20 °C and 40 °C, respectively:(1)TBHQ and TP had the same effect on rapeseed oil and soybean oil stored at 20 °C and 40 °C,they could restrain the auto-oxidation of oil welly.(2)PQQ at 0.8 mg/kg could restrain the auto-oxidation of rapeseed oil stored at 20 °C and 40 °C welly. TP at 200 mg/kg had no good effect on rapeseed oil when it was stored at 20 °C, however when it was stored 40 °C, TP 200 had opposite effect.(3)GPC at 100 mg/kg had good effect on soyben oil when it was stored at20 °C, however when it was stored at 40 °C, GPC 100 had bad effect.Experiment 2:Comparetive study of diffierent kinds of antioxidant in old laying hensFive hundred and seventy six Roman laying hens aged 64-weeks-old were randomly assigned to 8 treatments with 6 replicates of 12 hens each. Laying hens in control group were fed a basal diet, and those in TBHQ, TP, VE, PQQ, GPC were fed a basal diets supplemented with 200 mg/kg TBHQ, 200 and 400 mg/kg TP, 85 mg/kg VE, 0.08 mg/kg PQQ, 50 and 100mg/kg GPC, respectively. The results showed that:(1)antioxidant could restrain the decrease of laying rate for old laying hens. The trend was as follows: TP 200>GPC 50>PQQ >TP400>GPC 100>VE>TBHQ.(2)egg quality: TP 400,PQQ,GPC 50 could restrain the decrease of haugh unit compared with contrast, GPC could increase the eggshell thickness significantly compared with TBHQ and VE, TP200 could significantly increase the eggshell strength compared with contrast and VE, TP200 could significantly increased the egg length compared with all groups at 8 weeks(P<0.05). Other index had no significant difference(P>0.05).(3)plasma antioxidant: TP supplementation level at 400 mg/kg, VE supplementation level at 85mg/kg and GPC supplementation level at 50 mg/kg resulted in significantly decreasing the content of Malondialdehyde(MDA), TP supplementation level at 400 mg/kg and VE supplementation level at 85 mg/kg resulted in significantly increasing the activity of total antioxidant capacity(T-AOC)(P<0.05), other groups had no significant difference(P>0.05).VE supplementation level at 85 mg/kg and GPC supplementation level at 50 mg/kg resulted in significantly increasing the activity of total superoxide dismutase(T-SOD) compared with other groups, and TBHQ and PQQ significantly increased compared with contrast, TP and GPC 100(P<0.05).(4) liver antioxidant: TBHQ supplementation level at 200 mg/kg, TP supplementation level at 400 mg/kg, VE supplementation level at 85 mg/kg, GPC supplementation level at 50 mg/kg could significantly decrease the content of MDA; TP supplementation level at 400 mg/kg, VE supplementation level at 85 mg/kg and GPC supplementation level at 100 mg/kg resulted in significantly increasing the activity of T-AOC(P<0.05); TP supplementation level at 400 mg/kg, GPC supplementation level at 50mg/kg could significantly increasing the activity of T-SOD(P<0.05).(5)egg yolk antioxidant and cholesterol content: Dietary PQQ significantly increased the activity of T-SOD in egg yolk compared with other groups; other groups also had a little increase compare with contrast. The content of MDA in egg yolk significantly decreased in experiment groups(P<0.05). Dietary GPC significantly decreased the content of cholesterol in egg yolk(P<0.05) compared with the contrast and VE group.(6) plasma lipid metabolism and biochemical parameters: the content of cholesterol and triglyceride in plasma had no significant difference in all groups(P>0.05); VE supplementation level at 85 mg/kg and GPC supplementation level at 100 mg/kg resulted in significantly decreasing the content of L-DLC,GPC 100 was also significantly lower than TBHQ, TP, PQQ and GPC 50(P<0.05).Conclusion:1) The ability of antioxidant activity and scavenging DPPH, about this five kinds of antioxidants, were as follows: TP>GPC>TBHQ>VE. PQQ could not react.2) 100 and 200 mg/kg TBHQ and 400 mg/kg TP had the best effect on restraining theoxidant of oil. And 100 mg/kg GPC had good effect when the soyben oil stored at 20 °C; 0.8mg/kg PQQ had good effect on the rapeseed oil when it was stored in this two kinds of situation; but VE could not help this two kinds of oil in this experiment.3) 400 mg/kg TP had the best effect on improving the antioxidant activity in old laying hens, and 85 mg/kg VE, 50 mg/kg GPC followed it. PQQ as a new antioxidant, it could significantly restrain the decrease of laying rate and haugh unit in old laying hens, but had no significant effect on antioxidant.
