Polygonum ciliinerve(Nakai) ohwl is the root of Antenoron filiforme,and possess clearing heat-alleviating pain,hemostasis,acesodyne,anti-bacteria,anti-virus,antioxidant,et al,which is also the traditional seven medicine of Taibai Moutain area, Shaan Xi,China.Polygonum ciliinerve(Nakai) ohwl polysaccharides(PCP)is one of the effective extracting components and possess anti-bacteria,anti-virus,antioxidant activities.To study the immune effect of its sulfation(perpared by our research office already) on chicken immuned with Newcastle Diease vaccine,the follow experiments were conducted. PS group,BC control group, PCP group and three different substitution degree sPCP group with high(8mg/mL) and low(4mg/mL)dose were seted in this test and after the first baccined,PCP and sPCP group were injected with corresponding medicine,PS group and BC control group were injected with equalSterile Saline Solution,once a day for 3 successfully days,BC control group without deals.On days of 7th(D7)、14th(D14)、21th(D21)、28th(D28)、35th(D35)and 42th(D42) after the first vaccined,peripheral blood were collected and serum were separated.β-microdeterminatio method of blood coagulation test were conducted to test the ND-HI antibody titer, the concentration of IL-2、IL-6 and IFN-γ were determined by enzyme linked immunosorbent assay(ELISA),immune organ index were measured,and peripheral lymphocyte proliferation were tested by MTT method.Results are as follows:First, at the each time point, the antibody titer of PCP group were higher than that of PS and BC group,but no significant difference;The HI antibody titer of sPCP groups were significantly higher than other groups(P < 0.05), and the sPCP1 H group showed the best effect.Second, the thymus index at determinated time points in sPCP group were significantly higher than that of PS and BC group, but the spleen and bursa of fabricius index were higher than that of PS and BC group only on D28、D35 and D42(P < 0.05). Immune organ index of PCP group were higher than that of control group with significant difference only after the D28(P <0.05). sPCP groups also can significantly improve the Newcastle disease vaccine immuned chicken serum IL-2, IFN-γ and the content of IL-6 in high dose and the sPCP1 group showes the best effect.Third, at each time point, low doses of PCP can significantly promote T lymphocyte proliferation(P < 0.05);All sPCP can significantly promote the chicks peripheral blood T lymphocyte proliferation(P < 0.05), and the low dose s PCP1 group was the best.Conclusions: sPCP and PCP cooperated Newcastle Disease vaccine can improve and maintain the HI antibody titer of Newcastle disease vaccine immuned chicken, and had certain relationship to the degree of substitution and dose;sPCP and PCP can improve the Newcastle Disease vaccine immuned chicken organs of thymus, spleen and bursa of fabricius index and promote the development of chicken immune organs,sulfated modification can also improve the Newcastle disease vaccine immuned chicks the content of IL-2, IFN-γ and IL-6in serum, high dose sPCP1 effect is the best. PCP and sPCP can promote the Newcastle disease vaccine immuned chicken peripheral blood lymphocyte T cell proliferation, sPCP immune enhancement effect is superior to the PCP and sPCP at low dose effect is best. The results showes that sulfated modification can significantly improved PCP immune enhancement activity, improve the immune effect of Newcastle disease vaccine to chicken,indicates that sPCP can be used as a candidate for a new type of immune enhancer.
