The aim of this study was to explore the expression changes of fat metabolism related genes and the relationship between Villin2 and β-catenin in the process of fat metabolism by studying the effects of lithium chloride on differentiation and the expression of Villin2 and β-catenin in 3T3-L1 preadipocytes.In the study, the first experiment was to detect the state of 3T3-L1 preadipocytes differentiation by oil red O staining method. Li Cl’s concentration was 0m M, 10 m M, 20 m M and 30 m M. The result suggested that 20 m M Li Cl concentration was the better treatment concentration of inhibiting adipocyte differentiation. The second experiment was to detect the expression of fat metabolism related genes,Villin2 and β-catenin in 3T3-L1 preadipocytes on the 8th day of differentiation. The experiment was divided into control group and 20 m M Li Cl treatment group. The results suggested that 20 m M Li Cl concentration can regulate the expression of fat metabolism related genes and improve the expression of Villin2 and β-catenin gene on the 8th day of differentiation. The third and forth experiments were divided into control group and 20 m M Li Cl treatment group and to detect the expression of fat metabolism related genes, Villin2 and β-catenin in 3T3-L1 preadipocytes during differentiation-induction and maturity. The results suggested that 20 m M Li Cl concentration can regulate the expression of fat metabolism related genes and improve the expression of Villin2 and β-catenin gene during differentiation-induction and maturity.In conclusion, the study results showed that 20 m M Li Cl can inhibit 3T3-L1 adipocyte differentiation and can regulate the expression of fat metabolism related genes, FAS, LPL, ACC, PPARγ, c EBPα, SREBP1 c, GLUT4 and AP2. 20 m M Li Cl can enhance the expression of Villin2 and β-catenin not only during differentiation-induction, but also after differentiation-induction. Moreover, Villin2 had a significant correlation relationship with β-catenin. Villin2 participates in Wnt/β-catenin signaling pathway.
