Embr yo implantation plays a crucial role in mammalian repr oduction process, in which embryo contact with the maternal endometr ium and establis h structural connnection, in or der to achieve mater ial exchange processes. Goat(Capra hircus) belong to ruminants, whose embr yo implantation is a surface implantation, instead of invading into the uterus stromal, the trophectoderm cell fus e with endometr ial epithelial cell to for m cotyledonary placenta. Embr yo implantation ref ers to interaction between trophectoder m and epithelial c ells, the f eature of surface implantation make goat a useful model in study of inter action between maternal-fetal interf ace. As one of the wor ld’s oldest species, goat is an important component of her bivorous livestock, plays an importatnt r oal in the dairy, meat, f iber and other industr ies. Moder n embryo engineer ing technology has been widely used in br eeding of f ine var ieties, but the embr yos oper ated have a lower survival r ate when compar ed w ith the embryos natural developed, most of th e failur es occur dur ing implantation. Embryo implantation requires a complex inter action between the embryo and maternal uter us, environment of uter us is critical for implantation and even successful pr egnancy. Theref ore, study of genes express in goat uterus dur ing implantation is able to provide theoretical bas is for impr oving ability of implantation in goat.Transcriptome ref ers to the sum of all RNA tr anscr ibed in a particular tissue or cell in a certain state, inc luding m RNA and non-coding RNA, it can ref lect the expr ess ion and regulateion of genes from the whole. Studies on tr anscriptome analys is dur ing embryo implantation stage has progress in sheep and cattle. As the whole genome sequence of goat has been obtained,we perform uter ine tr anscr iptome s equenc ing and analys is dur ing implantation in goat. We except to reveal the changes in goat uterus transcr iptome dur ing implantation, screen out key genes that may play important role in embryo implantation, provide a bas is for understanding the mechanism o f embryo implantation in goat.Healthy Heilongjiang or dinary goat ar e feed under the same condition, uter ine tissue ar e collected on day 6(D6), day 16(D16), day 19(D19) of gestation, three goat are collected for each day to exc lude individual diff erences. RNA-Seq results showed that 1, 121 genes are differ ently expr essed between D6 and D16, in which 638 ar e up-r egulated, 483 ar e down-regulated; 2,555 genes are differ ently express ed between D6 and D19, in which 1,183 ar e up-r egulated, 1,372 are down-regulated; 452 genes ar e differ erntly expr essed between D16 and D19, in which 124 genes are up-r egulated, 328 genes ar e down-regulated. GO analys is reveale that diff erentially expr essed genes are most abundant in mammar y gland bud morphogenes is, d UMP catabolic p rocess, vir al envelope, CRLF-CLCF1 complex, neuromedin B receptor binding ter ms. Pathway revaele that differentially expr essed genes ar e most abundant in Nicotinate and nicotinamide metabolis m, RNA polymeras e, Pyr imidine metabolism, Glycosphingolipid bios ynthes is, Valine, leuc ine and isoleuc ine biosynthes is ter ms. We selection sever al genes to perform Quantitative Real-time PCR and In S itu hybr idization to ver if icate the RNA-S eq data, and validation results consistent w ith RNA-Seq results.Conc lus ion: Obtain candidate genes involved in goat embryo implantation, RNA-S eq data ar e ver if icated to be reliable through Quantitative Real-time PCR and In S itu hybr idization, which can be used for subsequent analys is, the RAN-Seq data lay foundation for subsequent stud ies.
