Atrazine, a widely used herbicide in the world, is a kind of important environmental pollutants, especially many wild birds and poultry in the environment. Liver is the main target organ where atrazine is accumulated and exerts toxicity. Atrazine also caused hepatocytes damage by excessive oxidative stress. Nrf2 plays a protective role against exogenous poison by regulating both drug metabolizing enzymes and antioxidant genes. Few reports about the relationship between Nrf2 and ATR in the body were reported, especially in the liver of quail.In the present study, quail liver as experimental object exposure to ATR. Then light microscopy, electron microscopy, q RT-PCR and western blot were used to detect the pathological changes of liver, antioxidant function, related indicators include CYP450, Nrf2, NQO1 and HO-1.The study indicated that:(1)Then the whole sequence of quail Nrf2 gene was cloned successfully first time, bioinformatics analysis showed that the protein was a non transmembrane, non secretory and non signal peptide, which was found in the cytoplasm and nuclear proteins; we successfully finished that the preparation of Nrf2 of rabbit polyclonal antibody against quail, the western blot method establishment to detective the protein level of Nrf2 in liver tissues.(2)ATR exposed to the liver tissues of the quail exhibited different levels of swelling, vacuole degeneration, necrosis, mitochondria swelling ridge fracture and endoplasmic reticulum vacuole, which plays a role of liver injury, and then affect the growth of quails.(3)The concent of CYP450 and the m RNA lelvel of CYP1B1, CYP2D6, CYP2C18, CYP2H1, CYP3A4, CYP3A7, CYP4V2 were increased caused by ATR in liver, disrupting the CYP450 enzyme system, which showed CYP450 enzyme played an important role in the prevention of liver injury induced by ATR.(4)The change of antioxidant functions related indicators, such as H2O2, MDA and GSH were increased, though GSH-PX, SOD and CAT were inhibited caused by atrazine in liver, which showed atrazine induced liver damage from excessive oxidative stress.(5)The m RNA level and the protein expression of Nrf2 were increased by ATR, the m RNA level of II phase enzyme NQO1 and HO-1 were also increase, indicates that ATR can activate Nrf2 signaling pathway by inducing oxidative stress, this is one of the important mechanisms of ATR induced liver injury in quail.
