Late blight, caused by the oomycete pathogen Phytophthora infestans, is the most devastating potato disease. Deployment of resistant varieties is an effective way to control this disease. We know that RPW8.1 confers broad-spectrum resistance to fungi pathogens that cause powdery mildew, and also enhances resistance to Hyaloperonospora arabidopsidis that cause downy mildew. RJPW8 protein is an atypical R protein (no NB-LRR structure). It specifically targeted to the extrahaustorial menbrane (EHM) and involved in enhancing the formation of a callose encasement of the haustorial complex and in onsite accumulation of H2O2. RPW8.1 gene also activates SA singal pathway during pathogens attack host cells.In this study we obtained 25 transgenic potato lines expressing RPW8.1 by the agrobacterium-mediated transformation. After infection with Phytophthora infestans, RPW8.1 can’t mediated resistance to Phyto- phthora infestans in potato. The expression of RPW8.1 was increased after infection with Phytophthora infestans. In SA signal pathway, key genes EDS1、 PAD4 and WRKY1 were significant up-regulated in transgenic lines, the transcripts level of these genes were continuously increased after inno-culation with Phytophthora infestans. No obvious expression change in JA and ET signal pathway between control and transgenic lines, while key genes in ET signal pathway were up-regulated after innoculation.
