Moso Bamboo is the most important economic bamboo species, has a long cultivation history, growing wide area, high economic value and ecological benefits and so on. Bamboo shoots are important products of bamboo industry in Fujian province, in 2010, bamboo shoots yield of 690,000 tons,41.7% per cent of national production, and bamboo shoots. Bamboo shoot is an excellent vegetable food, rich in fiber and protein, with a high nutritional value and medicinal value. In different seasons it has bamboo spring shoots, rhizome shoots and bamboo winter shoots, and all three have many differences in their morphology and quality, and each have a developmental approach. Bamboo winter shoots is beginning by the bamboo Rhizome of lateral bud in the autumn, it have high yield and popularity with people that sets its taste. Some research showed that, in bamboo different period of shoots its nutrition material content has changes, general trend is bamboo winter shoots above other period, which may be regulated by their own genes, however the growth mechanism of bamboo shoots is unclear,thus this project from molecular biology level research bamboo shoots into shoots mechanism, project used improved of CTAB method and Tiangen reagents box extraction bamboo RNA, We combined PacBio sequencing and Second Generation Sequencing (SGS) technology on bamboo shoots bud, and rhizome bud and the lateral bud reverse recorded into of full length cDNA, through Illumina 2500PE125 instrument measuring sequence, each samples average get 5.267G of data, which analysis has rhizome bud and shoots bud are related gene 6,863 a, rhizome bud and lateral bud are related gene 5,592 a, lateral bud and shoots bud are related gene 7029.Analysis of COG_ONTOLOGY of lateral buds, rhizome bud and shoot bud, its main functions of differentially expressed genes, the results are as follows:(1) the lateral bud genes involved in cell division and chromosome segregation and of carbohydrate transport and metabolism of biological processes.(2) the rhizome bud on the expression of genes involved in cell division, chromosome variation, biological processes such as transcription and DNA replication and repair.(3) the bamboo shoots bud the expression of genes involved in carbohydrate transport and metabolism, cell membrane production and lipid metabolism-related processes.GOTERM_BP_FAT analysis of lateral buds, the rhizome bud and shoot, its main functions of differentially expressed genes, the results are as follows:(1) expression of lateral bud genes involved in DNA metabolism, cell cycle and photosynthesis.(2) the rhizome bud genes involved in DNA metabolism and cell replication process.(3) the bamboo shoots bud expression of genes involved in photosynthesis, mainly in response to non-biological stimuli (such as photosensitive), there are also secondary metabolic processes.KEGG_PATHWAY analysis of lateral buds, the rhizome bud and shoot their metabolic pathways of differentially expressed genes, the results are as follows:(1) the lateral bud significantly expressed genes is primarily involved in the regulation of photosynthesis, DNA replication, and pyrimidinyl metabolism pathway.(2) the rhizome buds on main upregulation of genes involved in the regulation of DNA replication, DNA mismatch repair, pyrimidine or purine metabolism and chromosome by homologous recombination pathway.(3) the bamboo shoots bud expression of genes involved in photosynthesis, mainly in response to non-biological stimuli (such as photosensitive), there are also secondary metabolic processes.PIR_SUPERFAMILY analysis of lateral buds, the rhizome bud and shoot the differential expression of proteins, the results are as follows:(1) the lateral buds a significant expression of proteins is chlorophyll a/b binding protein, PIRSF esterase and Annexin group.(2) the rhizome bud significant expression of proteins include cell cycle related protein,H4 histone family, type of receptor-like kinases and kinase-associated protein.(3) on the bamboo shoots bud significantly expressed genes encoding proteins areXanthine dehydrogenase, cytochrome P450 family protein, α-crystallin-related smallheat-shock protein and protein in response to Abscisic acid category; bamboo shoots bud on the rhizome buds significantly expressed genes encoding proteins with the chlorophyll a/b binding protein (CAB), Annexin I and esterase and lyase.This study discusses the endogenous mechanism of bamboo shoot of Phyllostachys pubescens as bamboo shoot helps to further systematic analysis of genes of related function.
