Ammonia oxidation of microorganisms is the key of catalytic nitrification, is the main bearing in the process of soil nitrogen transformation. On the basis of long-term fertilization on brown soil in ShenYang Agriculture University, using real-time PCR (qPCR)technology combine with the soil physical and chemical properties, this study aims to analyze influence of long-term rotation and fertilization on ammonia oxidation microorganisms abundance in brown soil, the prime results are as follows:(1)The significantly effect of different fertilizer treatments on soil physical and chemical properties. Long-term application of manure fertilizer to the soil pH, alkali solution nitrogen (AHN), available phosphorus (AP), available potassium (AK) and soil organic matter (SOM) and total nitrogen (TN) content increased prominently. The long-term fertilization of mineral fertilizer reduced the soil pH value, single nitrogen treatment with the minimum of soil pH, There are significant differences in different fertilizer treatments of soil ammonium nitrogen and nitrate nitrogen. It’s good for accumulating soil ammonium nitrogen and nitrate nitrogen to fertilize manure, soil pH value, available phosphorus and available potassium, soil alkali-hydrolysable nitrogen, soil organic matter, soil total nitrogen and ammonium nitrogen and nitrate nitrogen content is a significant positive correlation. Show that long-term application of organic fertilizer helps maintain soil quality, and unbalanced fertilization makes soil conditions worsen.(2)Long-term rotation and fertilization conditions, the number of ammonia-oxidizing archaea is obviously higher than ammonia-oxidizing bacteria. The abundance of ammonia oxidizer under organic fertilizer treatments are significantly more than CK and singal chemical fertilizers treatments, the most number of ammonia oxidizer is M2, nitrify fertilizer treatments are the least.Manure(M2) treatments and nitrify fertilizer treatments(N1,N2) have a significant influence in ammonia oxidizer in brown soil. These all make it clear that long-term application of organic fertilizer can prominently improve the quantity of ammonia oxidizer.(3)The abundance of ammonia oxidizer archaea in all treatments are the same gragually increasing regular with them changing in plant growth stage. However, it is not the same on ammonia oxidizing bacteria in different growth stage. M of the number of ammonia oxidizer bacteria is firstly increasing then decreasing, converging stable at last. No fertilizer and singal chemical fertilizers treatments keep stable in pre-sowing, seedling and jointing three growth stage, but in tasseling and harvest stage do not find ammonia-oxidizing bacteria amoA gene, it concluded that the application of organic fertilizer to help maintain soil microbial communities ammonia oxidation stability.(4)By RDA analysis, long-term rotation and fertilization conditions, the factor of soil physicochemical properties on ammonia-oxidizing organism abundance in brown soil, there are soil pH, alkali solution nitrogen, available phosphorus, available potassium, soil organic matter and total nitrogen is a significant positive correlation. The environment factors affecting of the abundance of AOB with the corn growth period are soil pH, alkali solution nitrogen, soil organic matter, nitrate nitrogen, dissolve organic carbon, total nitrogen, but the abundance of AOA with the corn growth period environment factors are soil pH, soil moisture, soil organic matter.
