Toxoplasma gondii(T. gondii) is an intracellular parasitic protozoan and found in all warm-blooded vertebrate animals, including mammals, humans and birds. Toxoplasmosis is a kind of infection caused by T. gondii, which is a worldwide popular zoonotic parasitic diseases, posing a great threat to human health, also causing serious damage and great economic losses for livestock husbandry development. And for now, there isn’t an ideal remedy specifically for this disease. So we are looking for a drug with good efficacy, low toxic and less side effects. It’s reported that Lumefantrine is effective to malaria recently. Owing to the way of invasion and proliferation in Plasmodium and T. gondii is familiar, so Lumefantrine was chosen for the study of the inhibition in T. gondii by in vitro and in vivo experiments, and exploring the treatment effect and mechanism.Lumefantrine has no toxic action by MTT detecting and Giemsa staining in vitro. The results of FCM and MTT reveal that Lumefantrine can inhibit both invasion and proliferation of T. gondii, and which has a better impact on the inhibition of proliferation.This study has established the mouse models of acute T. gondii infection in vivo successfully, and using Lumefantrine for treatment. The content of T. gondii in different organs was tested by PCR and Real-time PCR and the change of cytokines in mice was detected by ELISA. The results show that Lumefantrine own the ability of prolonging the lifetime and reducing the mortality of mice. What’s more, the accounts of T. gondii in different organs from two drugs, Lumefantrine and Sulfadiazine, are less than the control. Furthermore, the efficiency of Sulfadiazine is worse than Lumefantrine which can change the amounts of cytokines to inhibit T. gondii.In this study, it’s indicated that Lumefantrine could inhibit the invasion and proliferation of T. gondii infection and has a greater effect on inhibiting proliferation. And Lumefantrine could prolong the lifetime of mice in acute T. gondii infection. All in all, this study will provide some available values and theoretical basis with the research of renewable anti-T. gondii drugs.
