Broccoli is a functional health-vegetable, and it contains aboundant sulforaphane (the best anti-cancer substance in vegetables). Methyl jasmonate is a kind of plant hormones, which has significant effect on the synthesis of glucosinolate-the premise of sulforaphane and activity of myrosinase. The selenium and sulfur should be competed in plants, but selenium can regulates the glucosinolate and sulforaphane synthesis in broccoli. In this paper, we analyzed the sulforaphane content and the myrosinase gene expression character of broccoli seeding and curd, which treated with methyl jasmonate and selenium. Aim to explore the optimum application concentration of methyl jasmonate and selenium to improve the content of sulforaphane in broccoli. It will provide the basis for the high sulforaphane broccoli production, and raw material production for large-scale sulforaphane industry. The results showed as follows:1 Exogenous application of methyl jasmonic acid could improve the broccoli sulforaphane content siginificantly. The sulforaphane content in broccoli seeding reached to 3000-4000μg· g-1, which 0.4 to 4.5 times higher than the control,3 d after 60-100μmol· L-1 methyl jasmonate treatment. The sulforaphane content in broccoli curd was 400-1200μg·g-1, which 2-6.7 times higher than the control,5 d after 20 μmol · L-1 methyl jasmonic acid treatment.2 Exogenous application of selenium could improve the broccoli sulforaphane contene significantly. The sulforaphane content in broccoli seedling reached to 1000-10000μg· g-1, which 2 to 3 times higher than the control, with 0.8-1.6 mg· plant-1 selenium treatment. The sulforaphane content in broccoli curd was 600-2500μg· g-1, which 1 to 5 times higher than the control, with 0.4-1.6 mg · plant-1 selenium treatment.3 Methyl jasmonic acid could significantly improve MY gene expression. The relative expression of MY gene in broccoli seeding enhanced 3-200 times than the control, with 20-60μmol· L-1 methyl jasmonate acid treatment. The relative expression of MY gene in broccoli curd was 0.5-14 times than the control, with 100μmol· L-1 methyl jasmonic acid treatment. But there is no significant correlation between gene expression and sulforaphane content.4 Exogenous application of selenium could improved the relative expression of MY gene significantly. The relative expression of genes in broccoli seeding was 2-100 times than the control, with 0.8-1.6 mg · plant-1 selenium treatment. The relative expression of MY gene in broccoli curd was 2-5 times than the control, with 0.8-1.6 mg · plant-1 selenium treatment. MY gene expression and sulforaphane correlation analysis shows that there is no significant correlation. |
