| Flammulina velutipes is one of the main cultivar in factory. As the popularization of cultivation techniques in factory, the number of mushroom residues of Flammulina velutipes increased greatly, therefore, the recycle study of the mushroom residues has become a focus. This paper studied the recycle of the mushroom residues which discarded in factory, extracted Flammulina velutipes residue polysaccharide by hot water extraction, optimized the process parameters; separated and purified the polysaccharide of enoki mushroom residues by macroporous adsorption resin; preliminarily determinated the α-glycosidase enzyme inhibition effect; And determinated its effect on plant virus disease prevention by using enoki mushroom residue polysaccharides extract as raw material. The purpose of this paper is to provide reliable data and foundation for industrial production of polysaccharide for large-scale use of enoki mushroom residue. The main research conclusions are as follows:1. Studied polysaccharides extraction method of enoki mushroom residue, because raw material sources and polysaccharides extraction dosage of enoki mushroom residue is large, so extracted the polysaccharide by the traditional hot water extraction. This method is simple and easy to operate, convenient for management. This paper studies the system including various factors influenced the extracted rate of polysaccharide, optimized the process parameters. The results showed that the optimum conditions of hot water extraction as: leaching temperature 90℃, leaching time of 6 hours, material liquid ratio 1:40. Through the analysis of variance in the needle mushroom extraction experiments, the size of the factors affecting the order, time or greater than the rest factors,such as the acuity of material liquid, temperature.2. Using five kinds of macroporous adsorption resin (ADS-17, AB-8, HPD-750, NKA-9, D-101) to separate and purify polysaccharide of enoki mushroom residue, D-101 is best in the adsorption and desorption effect. Several important factors in the process of adsorption and desorption were studied, the results showed that the best condition of adsorption is:material liquid concentration was 4.00 mg/mL, column velocity was 2 BV/h, column volume of up to 3 BV; 30% ethanol for elution, velocity 3 BV/h,6 BV elution liquid product.3. The inhibiting effect of α-glycosidase enzymes of noki mushroom residue polysaccharide were simply analyzed. Its IC50 is about 2.85 mg/mL, while acarbose IC50 is about 7.5 mg/mL.From the analysis on the results, needle mushroom polysaccharide material slag for alpha glycosidase enzyme inhibitory effect than acarbose positive control group, the result is about 2.5 times the effect of acarbose.4. Enoki mushroom residue as raw material, extracted enoki mushroom residue polysaccharide, and then measured its prevented and cured effect on plant virus disease. Studies have shown that it could prevent and control the infection of virus when the crops (tobacco, tomato, cucumber) were sprayed enoki mushroom residue polysaccharide with the dilution between 1000 and 4000. The control effect was up to 76.0% or more, especially when the dilution is 1000 times, the control effect was 81.9% or more. |
