The in vitro potato plantlet technology is the critical technique of potato breeding and multiplication; to obtain vigorous seedlings in vitro is the fundamental step in potato seedling production. After several times of successive transfers of culture during rapid propagation of in vitro potato in vitro plantlet, long-term use of test tube seedlings will cause several degradation phenomenon such as the plants becomes slim, the leaves turns yellow, growth vigor becomes weak. Only when we cultivate potato test tube seedling with stout stem and well developed root system, can we promote the well development of potato seed breeding system efficiently and provide enough virus-free seed potato for production.In this study we applied Chunshu5 and D613 as our experimental materials to study cultivating strong plantlets and transplantation. We also investigate the effect of plant growth regulators on the transplantation of potato test tube seedlings in order to provide a synthesis technique which can obtain potato seed breeding. The major experimental conclusions were as follows:1.Through the use of plant growth regulator PP333、CCC、B9、6-BA, the Chunshu5 in vitro plantlet grew well. This means that these plant growth regulators have a role of promoting the test tube seedling to vigorous growth. By the comparison analytic method, we concluded that Chunshu5 in vitro plantlet strong plant culture can be obtained with 10mg/L CCC or 5mg/L B9 or 10mg/L or B9.2.The survival rate of transplantation of Chunshu5 test tube seedling has been significantly improved after treated with plant growth regulators compared to blank control. After analytical comparison with survival rate, number of tubers, acceptability, ratio of big tuber, it was all suitable for Chunshu5 cultivation and transplantation treated with 10mg/L CCC、5mg/L B9、10mg/L B9.3.Through the use of plant growth regulator PP333、CCC、B9、6-BA, the growth of D613 in vitro plantlet way relegated differently. By the comparison analytic method, we concluded that it performs well with 2mg/L PP333 10mg/L B9 in D613 in vitro plantlet strong plant culture.4.The survival rate of transplantation of D613 test tube seedling was affected when D613 test tube seedling was treated with plant growth regulators. PP333 treatment of 0.2 mg/L and 0.5 mg/L 6- BA processing, have reduced the D613 tube seedlings transplanting survival rate. After analytical comparison with survival rate, number of tubers, acceptability, ratio of big tuber, only 10 mg/L B9 treatment can be applied in D613 in vitro plantlet transplantation.In a word, along with the different potato genetic background, it shows obvious difference when choosing different plant growth regulators, choosing different concentration. We must be must based on the varieties of potato, cooperate with processing with suitable type and dose of regulator, can we achieve strong seedling of plantlet in vitro and promote the survival rate of transplanting, number of tubers and improve the quality of potato products.
