Gibel carp come from Dafeng Huachen Fishery Cooperatives and Dafeng Jinlu Fishery Cooperatives. Dafeng Huachen Fishery Cooperatives: stocking model, Mixotrophic Culture of Gibel carp and grass crap. Dafeng Jinlu Fishery Cooperatives: stocking model of Sick Gibel carp, Mixotrophic Culture of Gibel carp and Ictalurus Punctatus, stocking model of Healthy Gibel carp, monoculture.Test 1 Studies on the Cyprinid herpesvirus 2 and Ultrastructure and histopathology of Gibel carp(Carassius auratus gibelio) Infected with Cyprinid herpesvirus2(Cy HV-2)The ultrastructural pathology and histopathology was observed in mesonephros, spleen, pronephros, hepatopancreas, intestine, and gill of gibel carp(Carassius auratus gibelio) with hematopoietic necrosis by transmission electron microscopy and optical microscope in order to analyze cyprinid herpesvirus 2(Cy HV-2) morphology, distribution, occurrence process and damage of tissues and organs in gibel carp with hematopoietic necrosis. Cy HV-2 was detected in the all organizations, and had four different maturity periods and diameters, 65-90 nm in DNA core, 90-180 nm in empty capsid, 90-180 nm in solid nuclear capsid, and 170-220 nm in enveloped viral particles. There was a large number of Cy HV-2 virus in mesonephros and spleen, with less Cy HV-2 virus in pronephros, hepatopancreas, intestine and gill. Electron microscopy observation revealed that immune cells were infected with Cy HV-2, which may lead to immune function change. Ultrastructurally, marginalized nucleus, nuclear chromatin degeneration, disrupted nuclear membranes were observed in the phagocytes in above organs, including the swollen markedly mitochondria and pieces of some dissolved cells. Histopathological changes of gibel carp have been studied that cell lysis, immune cell infiltration, serious damage to the immune and hematopoietic systems in head kidney and spleen; Glomerular atrophy, tubular epithelial cell shedding, interstitial vacuoles, immune cell infiltration in kidney; villi hyperplasia, the length of the villi and microvilli had significantly reduced, the submucosa had Significantly thickened in intestinal; Liver cells appeared to dissolve, central venous damaged in hepatopancreas; Capillary damage, blood vessel rupture or expand, RBC overflow, the cell had significantly reduced in the base of gill lamella. The results showed that Cy HV-2 can be diagnosed by characteristics of the virus particles via PCR and electron microscopy. The Cy HV-2 is observed in major organs of unhealthy gibel carp, and primarily replicated and assembled in the nucleus, and the n obtain the envelope in the cytoplasm.Test 2 Studies on the variation of blood biochemical indices and histopathological in gibel carp(Carassius auratus gibelio)infected with Cyprinid herpesvirusⅡIn order to study on the variation of blood components and functions and to discussion the pathogenesis of gibel carp with Cyprinid herpesvirusⅡ. The experiment group1(H) which collect virus carriers fish from ponds of never pathogenesis, the experiment group2(IH) which collect in the early stage of a disease fish from ponds of being sick, the experiment group3(I) which collect in the later stage of a disease fish from ponds of being sick. The three different pathological conditions gibel carp as samples in Dafeng city, Jiangsu province. We found that Compared with the H group, IH and I group, apart from lymphocyte percentage was significantly increased(P<0.05), neutrophilic percentage, monocyte percentage, erythrocyte count, hemoglobin, platelet count, platelet hematocrit were significantly decreased(P<0.05), Compared with the IH group, lymphocytes percentage of I group was significantly increased(P<0.05) while neutrophils percentage was significantly decreased(P<0.05), other indexes had no significant change, after determined blood indexes of three groups. Compared with the H group, IH and I group, apart from AST/ ALT, ALB/ GLB were significantly increased(P<0.05), while CHE, ALP, TBA, TG, HDL/LDL were significantly decreased(P<0.05), Compared with the IH group, AST, AST/ ALT, ALB, ALB/ GLB, of I group were significantly decreased(P<0.05), while CHE, ALP were significantly decreased(P<0.05), the rest had no significant changes, the content of TBA was decreased by 38 times, decreased significantly. By scanning observation, erythrocyte of H group surface smooth and complete, while erythrocyte of IH group and I had some injuries; compared with H group, hepatopancreas and intestinal tissue of IH and I group structural damage, and with different carp increase the severity of the illness aggravated.
