In order to understand the variation situation of duck hepatitis A virus Type Ⅰ,6 duck hepatitis A virus strains’ SD4、SD10、SD15、SD37、SD38、SD40 from Shandong province was inoculated in duck embryo allantoic cavity, the virus allantoic fluid was collected, identified and Froze for application. The VP1 Gene was sequenced, nucleotide homology was compared and evolutionary tree was drew with fourteen duck hepatitis A virus strains announced in GenBank by MEGA5. The results showed that, the homology of SD4 and ten duck hepatitis A virus Type 1 strains’ VP1 gen was 84.7% to 91.0%, the homology of SD10 and ten duck hepatitis A virus Type 1 strains’ VP1 gen was 86.5% to 92.2%, the homology of SD15 and ten duck hepatitis A virus Type 1 strains’ VP1 gen was 84.9% to 90.4%, the homology of SD37 and ten duck hepatitis A virus Type 1 strains’ VP1 gen was 84.8% to 90.3%, the homology of SD38 and ten strains duck hepatitis A virus Type 1 VP1 gen was 92.2% to 98.0%, the homology of SD38 and ten strains duck hepatitis A virus Type 1 VP1 gen was 85.9% to 91.8%. The homology of 6 test strains and duck hepatitis A virus Type 2 and Type 3 was 62.5% to 71.6%, which was lower than Type 1. The homology of SD38 and ten strains duck hepatitis A virus Type 1 VP1 gen was higher than other test strains.Meanwhile, the VP1 gen of SD38 strain was expressed fractionally. High antigenicity solvable protein pCold-TF-1Q, pCold-TF-1Z, pCold-TF-1H was obtained successfully, the fractional expressed protein was purified. The test layed foundations of building ELISA detection method.
