To investigate the prevalence of the Bluetongue disease in Inner Mongolia, the major types of the Culicoide as spreading media, the main popular serotype of the Bluetongue disease and the Epizootic Haemorrhage disease in Inner Mongolia. In this essay, using morphology and molecular biology methods to identify the species of the Culicoide. Animal serums collected from different home feeding ruminants randomly in different regions of Inner Mongolia since 2013 for serum epidemiology investigation. Used virus neutralization test to identify the main popular serotype of the Bluetongue disease and the Epizootic Haemorrhage disease in Inner Mongolia, selecting the samples’ which inhibition rate are over 60%. Used the monitoring animals’ EDTA total blood to isolate virus, and used the methods of the Antigen Capture ELISA and RT-PCR and Real-time RT-PCR to detect the Bluetongue virus.The results showed that the Bluetongue virus positive rates of samples were 10.82% in 2013,11.75% in 2014,11.27% in 2015. In addition, the Bluetongue virus infection was 8.76% in sheep,8% in cashmere goats, and 23% in bovine in 2014; and 10.25% in sheep, 15.56% in cashmere goats, and 0% in bovine in 2015. The Epizootic Haemorrhage disease virus positive rates of samples was 1.43%, including 2.46% in sheep,1.15%in cashmere goats,0% in bovine in 2015. The Bluetongue virus positive rate remained at 10% to 12%, little change, but has a tendency to increase. The wild ruminants’ Bluetongue virus positive rate was 3.67%, which include elks, deer, antelopes and blue sheep. The wild ruminants Bluetongue virus positive rate is lower than the home feeding ruminants, and the sheep is more susceptible than other ruminants. The Bluetongue virus-7,10,23 and the Epizootic Haemorrhage disease virus 6 are the main popular serotypes in Inner Mongolia. Through the classical separation method, no Bluetongue virus has been separated in Inner Mongolia. By mathematical morphology method combined with molecular biolgy methods, identified the major types of the Culicoide, such as Culicoides putianensis, Culicoides circumscriptus, Culicoide imicola.These data demonstrated that the prevalence of the Bluetongue virus is widely popular in Inner Mongolia. Therefore, understanding the distribution and prevalence of those two viruses is meaningful and necessary to strengthen the detection and prevention of those two viruses.
