Exogenous Nutrients-caused Development Of Psammosilene Tunicoides Hairy Root

Abstract/Summary:
Psammosilene tunicoides is Caryophyllaceae family, genus of P. tunicoides, which is the major ingredient of “Yunnan Baiyao”, one of the most famous Chinese Medicines. Now, the demand for P. tunicoides resources in the market of herbs increases remarkably, meanwhile, the population of wild P. tunicoides resources is facing to be exhausted. After successful induction and production in large-scale, the culture of P. tunicoides hairy roots could be an alternative to its wild resources, which will be a promising means to realize the sustainable exploitation of wild P. tunicoides resources.This study aims to characterize the growth state of P. tunicoides hairy root affected by modifications of vital factors, which include basic media, carbon sources, nitrogen source, phosphate ion, calcium ion and hormone, et al. First, the P. tunicoides hairy roots were cultured in different media with various formula of nutrient composition; After a period of culture, observation of P. tunicoides hairy root configuration was conducted, and then their dry weight of P. tunicoides hairy root were measured and analyzed. Finally, through evaluation of hairy root growth and changes in dry weight, the optimal formulation of nutrients suitable for P. tunicoides hairy roots growth was determined.The results showed that,(1) for solid culture, the preferred formula of culture media was demonstrated to be B5 + sucrose 30 g/L+ total nitrogen 60 mmol/L + NO3-/ NH4+ 2: 1+ phosphate ions 3.0 mmol/L+ calcium ions concentration 1.5 mmol/L+ 0.5 mg/L NAA +0.3 mg/L IBA.(2) for liquid culture, the preferred formula of culture media was demonstrated to be B5 + sucrose 30 g/L+ NO3-/ NH4+ 4: 1+ phosphate ions 0.7 mmol/L+ calcium ions 0.9 mmol/L+ 0.05 mg/L NAA+ 0.2 mg/L IBA, and the hairy roots of P. tunicoides exhibit properties of strong growth, heavy branching and maximal dry weight of 9.936 g/L.(3) the anti-bacteria activity of P. tunicoides hairy roots was also examined, and the results showed that the culture media had evident effect in this respect, however, the extract of P. tunicoides hairy roots performed strong inhibition on bacteria.Generally, the expected growth rate of P. tunicoides hairy roots was realized to some extent through optimization of nutrimental ingredients in the culture medium, by which both growth and biomass were at their peak. The above results would provide theoretical support for in vitro production of P. tunicoides hairy roots in large scale in the future.
