This study was designed to isolate, identify and biological characteristics of the Capripoxvrus from samples separated from sheep that were suspected to be infected by the (apripoxvirus on one farm of Hohhot and A Lashan Inner Monglia. We used cells of □ Marc-145 and Vero to separate and culture the objective virus and by the way of animal Regression experiment. observing histopathological slices、observing by transmission electron microscope. PCR identification to do the identification of the (apripoxvirus. Then we designed and synthesized a couple of special primers according to the P32 gene sequence of the Sheep pox virus published in the GeneBank. Meanwhile, we successfully did the sequencing and cloning for the P32 gene of the separated strains and the comparative analysis of homologies with other conferenced strains.The results showed that the two strains of the virus suspension in the LT. Marc-145 and Vero cells in continuous culture after 4 generations, can appear stable cytopathic effect. The clinical symptoms of test animals were the same as that of typical (apripowiriis in the animal regression experiment, the acidophilia inclusion body could be observed in the histopathological slices and spindle goatpox cells. The virus suspension made negative biopsy and ultra-thin, observed the typical pox virus particles under transmission electron microscopy. Two strains of the virus homology comparison with reference strains. Hohhot isolates and reference strains homology was 97.5%~ 99.4%. A la shan isolates and reference strains homology was 98.0%~99.9%, the homology between the two strains of virus isolates was 99.3% after the comparative analysis through DNASTAR Phylogenetic results show that the strain we had separated is the (apripowirus and we named it as CaPV strain Hohhot2015 and CaPV strain Al ashan2015...
