An iridovirus(tentatively named CQIV, Cherax quadricarinatus iridovirus) that was found and isolated from redclaw crayfish in Fujian in 2014. Healthy C. quadricarinatus, Procambarus clarkii and Litopenaeus vannamei were artificial infected with purified CQIV. To research the tissue/cell type-tropism of CQIV by knowing which types of cells can be infected by CQIV. TEM reveals that the tissue/cell type-tropism of the three crustaceans infected by CQIV showed a high degree of consistency. Phorocyte, hematopoietic cells and epithelial cells of gill were the major target cells, which can be infected and multiplied in the cytoplasm by CQIV. Most hemocyte can not be invaded and multiplied by viruses. Other cells such as the myocardial cell, muscle cell, oocyte, neurocyte, epithelial cells of midgut, absorptive cell(R cell) of hepatopancreas, blister cell(B cell) of hepatopancreas, fibrillar cell(F cell)of hepatopancreas and labyrinth cells of antennal gland were not infected by CQIV. At the same time, we took the larva of Litopenaeus vannamei as research object to learn the transmission that CQIV by the technology of Real-time PCR. The result of this study showed that the prawn can be infected by feeding meat with CQIV.Besides, we also took the larva of Litopenaeus vannamei and red swamp crawfish as research object to learn the transmission of WSSV by the infecting methords of soaking and feeding using the technology of Real-time PCR. In this study, in order to prevent the interactive infection, the experimental animals were strict fed separately. The result of this study showed that the prawn can be infected by the infecting methords of soaking and feeding. However the red swamp crayfish can be infected by the infecting methord of feeding only. Under the low feeding amount, feeding shell muscle with WSSV can be effectively infected the healthy and energy better red swamp crayfish than feeding muscle with WSSV only. we also found that the feeding amount has a positive correlation with the case of infection. At the same time, we took the redclaw as research object to learn the tissue/cell type-tropism of WSSV using the technology of TEM. The tissue/cell type-tropism of WSSV was roughly consistence with CQIV. In other words, connective cells, hematopoietic cells and epithelial cells of gill were the major target cells, which can be infected and multiplied by WSSV as well.
