| Porcine reproductive and respiratory syndrome is also well known as the "blue ear pig disease", which is harmful to the world pig breeding industry. It can cause reproductive obstacle disease and piglets respiratory diseases. The virus also can induce the immune suppression. Reducing immunity against other disease, there by leading to secondary infection or mixed infection with other diseases. How to control the disease effectively has become the central issue around the world. At present the immunity of PRRS in China mainly depends on inoculation with non-live vaccines, with little knowledge about the reaction of immune cells, therefore, the study on immune cells of pigs will provide new insights into the prevention and control the PRRSV.Dendritic cells as the strongest antigen presenting cells can only activate immune response of native T cell, but also play an important role in regulating immune responses, it also can induce immune to lerance in an certain circumstances. DC can secret the different types of cytokines to regulate the body’s immune response and induce the differentiation of T cells when stimulated by different antigens, which is crucial for the specific and non-specific immune response. Culturing the porcine dendritci cells in vitro infected by PRRSV, this study described the positon variation of antigens and the changes of cytokines secreted which provides the new direction for research on mechanism of PRRSV.(1)Dendritic cells were successfully separated and cultured in vitro from peripheral blood.(2)porcine reproductive and respiratory syndrome virus(PRRSV)had bred in Mac-145 cells using the virus amplification technology.(3) By using the advanced flow cytometry PRRSV in dendritic cells infected by PRRSV and expression of CD1 decreased, and expression of CD80 and SLA-II increased significantly.(4)The cell secretion of TNF-αã€INF-γã€IL-6 ã€tryptophan ã€IDO increased,and IL-10 decreased by ELISA in the process of cell culture.(5) The expression of indoleamine 2,3-dioxygenase(IDO) increased by Western blot after the cells had been infected by virus. It is concluded that DCs plays an important role in fighting against PRRSV infection mainly through the secretion of high levels of cytokines. |
