Avian influenza is caused by type A influenza virus from respiratory disease systemic sepsis to highly contagious acute infectious disease, chickens, turkeys, ducks, other poultry and wild birds can also be infected. The disease emerged in many countries and regions such as Asia, America, Europe, the poultry industry supplies caused enormous losses. Avian influenza can not only spread in poultry, seriously affect the aquaculture industry’s survival and development, but also can be transmitted from poultry to humans, a direct threat to human health, to human public health also has caused considerable harm. Recently, the country home and surrounding animal and human avian flu bird frequently happened, only domestic H5N1 avian influenza virus nucleic acid positive deaths reached five cases. Highly Pathogenic Avian Influenza World Organization for Animal Health has been classified as category A infectious diseases, and are included in the international Biological Weapons Convention, the list of infectious diseases of animals, our country will also be classified as a class of its animal disease.NS1 protein acts as the only non-structure protein that belongs to the type A of Avian influenza virus. NS1 protein contains 202-237 amino acids, which shows the some differential between the varies kinds of strain. NS1 protein has been transferred to the nuclear after been synthesized in the cytoplasmic matrix, as well as accumulate into the nuclear in the early period of infection. Later, these proteins have been gathered into the nucleolus. NS1 protein plays an important function on the virus replication, immune response and induction of the decline process. Because NS1 protein expresses at a high level in the early period of virus replication, which just exists inside the cell, could induce the body produce the special antibody against the NS1 protein. In recent years, both at home and abroad about the evolution of NS1 protein, structure, regulation of the pathogenicity of the virus and its applications to carry out an indepth study, through its interacting protein screening and identification, can be further revealed the A-type influenza virus-induced disease mechanism, as well as the avian flu virus caused the death of infected people and provide an important mechanism of the reference value.To screen and identify proteins that interact with the NS1 protein by means of T7-phage display system. The NS1 protein was expressed by prokaryotic expression and used as selectedmolecule to biopan the T7 human liver cDNA library. The selected positive clones were identified by DNA sequence and analyzed with BLAST program in GenBank. After BLAST in all positive clones, one protein Homo sapiens nucleolar and coiled-body phosphoprotein 1 (NOLC1)was found to interact with the NS1 protein.Screening of the protein, Human nucleolar phosphoprotein NOLC1, is a highly phosphorylated protein, showed the existence of granular nucleolus in the interphase nuclei, when cells in mitosis when there is, NOLC1 particles collapse of uniformly distributed to the cytoplasm, with RNAΙpolymerase interact and participate in the occurrence of nucleolus, rRNA gene transcription regulation of cell growth; happen inflammation; liver cancer occurrence and development has an important role in the regulation and control. This experiment for further study of the biological function of NS1 proteinto provide an important basis for the future development of anti-influenza drug to provide a new clue.
