In general condition, cough is an indispensable defensive reflex. Through discharge the sputum or the eyewinker in the respiratory tract to keep the respiratory tract cleaning and unobstructed, thus preventing infection formation; But if cough fierce continued, not only influence the daily life and work of patients, but also facility come into being other diseases. Cough is also a common symptom of diseases such as respiratory tract infection, asthma and chronic obstructive pulmonary disease (COPD), eosinophilic bronchitis (EB) and lung cancer. TZS is one of traditional Chinese medicine with common widely use, TZS is the root of Psudostellaria heterophylla (Miq.) Pax ex Pax et Hoffm.2010 edition of China pharmacopoeia recorded, TZS possesses nourishing qi to invigorate spleen, promoting fluid production to nourish lung, in clinical it commonly used in spleen asthenia and dry cough.This topic through the whole animal experiment by using two kinds of model in order to screen the effective polarity parts and activity chemical constituents, with a view to explore TZS in the material basis and mechanism of action for the further development of antitussive in new drugs provide an experimental basis and theoretical basis. This paper is divided into five chapters, The main contents of each chapter are as follows:The first chapter:Antitussive activity of Pseudostellaria heterophylla (Miq.) Pax effective polarity parts of the screenApply the cough model of citric acid induce guinea pigs, the cough frequency variation of guinea pig acquisition and processing system online record through Medlab biological connected breathing flowmeter on computer, thus screen the effective polarity parts of TZS extracts, different guinea pigs were gived different polarity parts of TZS extracts, by comparing the latent period and frequency variances intensity of cough (cough percent inhibition rate) in guinea pig before and after dose administration to determine the antitussive activity effective polarity parts of TZS. The experimental results showed that the total 85% ethanol extract, TZS water extract, ethyl acetate parts, polysaccharide extract, after dose administration reduced guinea pigs cough times and del latent period of cough in different degree. Including the groups of ethyl acetate part polysaccharide parts compared with model group with significant differences, the antitussive a of these two groups were obvious dose dependent, with repeatedly dosing can enhance anti(?) activity.The second chapter:Antitussive activity effective parts of chemical composition and mol combination group of studyPreparation of 85% ethanol extracts of TZS, which solvent extraction in turn by pet(?) ether and ethyl acetate, in this way getting petroleum ether extract, ethyl acetate extract respe(?) Using silica gel column and column chromatography method with chloroform and metha elution agents to separate the ethyl acetate effective polarity parts of TZS, using reserve-pha performance liquid chromatography (HPLC), Sephadex LH-20 gel column chromatogra separation and purification the distillate fractions, at the same time the compounds physicoc(?) properties and chromatogram structure were identificated. By this way, get eight m(?) compounds:β-sitosterol(Ⅰ),2-minali(Ⅱ),3-(4-hydroxy-3-methoxy- phenyl)propionic ac ursolic acid(Ⅳ), luteolin(Ⅴ),adenosine(Ⅵ),α-spinasterol-β-D-glucopyranosi-de(Ⅶ), daucost(?) The pharmacological activities of these eight monomer compounds have been reported in lite according to the relevant biological activity in our study, four compounds were choosed to stuⅣ,Ⅴ,Ⅷas the object of research, carry out the pharmacological research of the m(?) combination groups. The experimental results showed that monomer compositionⅠ,Ⅴh(?) antitussive activity, the antitussive activity ofⅠ+Ⅴwere strengthen thanⅠ,Ⅴsolo administration.The third chapter:antitussive mechanism of TZS extracts and effective components gro(?)Base on the first two chapters, further study using an animal experiment model of rat difficiency, antitussive activity extracts including ethyl acetate extract, polysaccharide par acetate extract and molecular combination of effective component parts intervention difficiency rat models general signs, pulmonary function, the immune function and lun pathology results change results were studied in this chapter. The results showed that TZS and active chemical composition can reduce generation and release of lung inflammatory cells chemotactic factors IL-8, TNF-a, GM-CSF, at the same time increase the anti-inflammatory cytokines IL-10 formation, and then control the growth of the disease, the mechanism may be that TZS inhibit lung inflammation, improve lung function.The fourth chapter:summary and prospectIn summary, we investigated the possible of material basis on antitussive activity of TZS, and look forward to the development and application prospect of TZS as new antitussive drugs.
