Objective To investigate the effect of TGF-β1、VEGF and IL-6expression in thetumor cells of the primary tumor and the lymph node metastase of patients withbreast cancer on Dendritic Cells (DC) in the primary tumor and the local lymphnodes.Methods The expressions of TGF-β1、VEGF and IL-6in cancer tissues of theprimary tumor and the lymph node metastases of52patients with breast cancer wereassessed by immunohistochemistry, using TGF-β1polyclonal antibody、VEGFmonoclonal antibody and IL-6polyclonal antibody. The numbers of DC in theprimary tumor and the local lymph nodes were also assessed byimmunohistochemistry using S-100antibody. The relationship between theexpressions of TGF-β1、VEGF and IL-6and the DC numbers was analyzed.Result1. In52cases of patient with breast cancer, the TGF-β1expression in the primaryfoci of40patients was positive, the positive rate was77.4%; the expression ofVEGF in the primary foci of48patients was positive, the positive rate was88.7%;the expression of IL-6in the primary foci of47patients was positive, the positiverate was90.6%.In23cases of patient with lymph node metastasis, the TGF-β1expression in theaxillary lymph node metastasis of13patients was positive, the positive rate was56.5%; the expression of VEGF in the axillary lymph node metastasis of22patientswas positive, the positive rate was95.6%; the expression of IL-6in the axillarylymph node metastasis of23patients was was positive, the positive rate was100%. 2. The average number of DC in the primary foci of52patients was8.31/hpf. Theaverage number of DC in379tumor-free lymph nodes of52patients was54.02/hpf.The average number of DC in242tumor-free lymph nodes of29patients with nolymph node metastasis was59.88±46.61/hpf. The average number of DC in137tumor-free lymph nodes of23patients with lymph node metastasis was42.04±33.69/hpf. The average number of DC in23tumor-containing lymph nodes of23patientswith lymph node metastasis was20.13±17.28/hpf.3. The TGF-β1、VEGF and IL-6expressions and the numbers of DC in the primarytumor of52patients was negatively correlated (P <0.05), the correlation coefficientwas-0.427、-0.327and-0.314respectively. The TGF-β1expressions in theprimary tumor of52patients and the DC numbers in379tumor-free lymph nodes ofthose patients was negatively correlated (P <0.05), the correlation coefficient was-0.144. TGF-β1expressions in the primary tumor of29patients with no lymph nodemetastasis and the DC numbers in242tumor-free lymph nodes of those patients wasnegatively correlated (P <0.05), the correlation coefficient was-0.188.4. The VEGF expression in axillary lymph node metastasis of23patients with lymphnode metastasis and the DC numbers in137tumor-free lymph nodes of thosepatients was negatively correlated (P <0.05), the correlation coefficient was-0.222.Conclusion1. The expressions of TGF-β1、VEGF and IL-6in tumor cells of Breast cancer maybe one of the reasons that decrease the number of DC in the primary tumor and thelocal lymph nodes.2. The local immune function is impaired with the number of DC decreased in theprimary tumor and the local lymph nodes in patient with breast cancer, resulting intumor cells escaped from immunity supervision. Objective To investigate the differences of expressions of ER、PR andCerbB-2in between the primary local foci and the lymph node metastases of patientswith breast cancer, and their clinical significance.Methods50cases of patient with primary breast cancer with lymph nodeinvolvement were treated surgically, their lymph node involvement were confirmedby pathological examination. The expressions of ER、PR and CerbB-2in cancertissues of the primary foci and the lymph node metastases were assessed byimmunohistochemistry, and their differences between the primary foci and the lymphnode metastases were statistically analyzed.Results The positive rates of expressions of ER and PR in primary foci werehigher than in lymph node metastases, their differences were significant (P<0.01). Thedifference between the positive rates of expressions of CerbB-2in primary tumors andlymph node metastases was not significant (P>0.05).Conclusion It is more resonable that the expressions of ER and PR in lymphnode metastasis of patient with breast cancer are used as indications of endocrinetherapy compare to those in primary foci.
