The Application In The Diagnosis And Prognosis Of Sepsis By Cytokine Antibody Chips

ObjectiveTo Screen the serum protein biomarker of patients with sepsis by cytokineantibody chipsMethodsIn this study, the antibody chip technology platform, the determination of the9cases of sepsis patients and79of the4cases of normal, healthy human serumcytokine signal value. Analyse the signaling values of these79cytokines using SAM(significance analysis of microarray) software. The33sepsis patients were devidedinto two groups:the dead group (18cases)and the cure group(15cases).Then analysethe signaling values of the two groups to find the serum protein biomarker ofprognosis in patients with sepsis by cytokine antibody chips.ResultsThe first part, by SAM (significance analysis of microarray) analysis of the sepsispatients and the healthy human:insulin like growth factor binding protein-1(IGFBP-1)，epidermal growth factor (HGF)，osteopotin，insulin likegrowth factorbinding protein-4(IGFBP-4)，interferon inducibleprotein-10(IP-10) andB-lymphocyte chemoattractant(BLC) were identified to be highly expressed in the sepsis while platelet-derived growth factor-BB (PDGF-BB)，brain-derivedneurotrophic (BDNF)，macrophage inflammatoryprotein-1β (MIP-1β)，interleukin-8(IL-8)，epidermalgrowth factor (EGF) and interleukin-1β (IL-1β)lowly expressed Cluster analysis of significantly expressed cytokines showed that thesepsis and the control formed distinctly separate groups. The sepsis patients and thevoluteer can be devided into two groups distinctly by cluster analysis.The second part,by SAM (significance analysis of microarray) analysis of the dead group and the curegroup:platelet-derived growth factor-BB (PDGF)、epidermalgrowth factor (EGF)、macrophage Inflammatory protein1beta (MIP-1beta)、macrophage inflammatoryprotein3Alpha (MIP-3Alpha)、Osteoprotegerin、monocyte chemoattractant protein-1(MCP-1)、interleukin-8(IL-8)、growth-related oncogene(GRO)、interleukin-10(IL-10) and interleukin-6(IL-6) were identified to be highly expressed in the deadgroup. The levels of IL-8and GRO protein are positive correlation with sepsispatients’life span by survival analysis.ConclusionAntibody chips demonstrate a significant change in sepsis patients, screeningthe protein biomarker for rapidly diagnosing sepsis and prognosis is feasible.
