Objective (1) To investigate the correlation of XIAP Livin and Caspase-3protein inhuman gastric cancer tissues and its correlation with clinical and pathologicalcharacteristics.(2) To investigate the role of XIAP Livin and Caspase-3protein in theoccurrence and prognosis of human gastric cancer tissues, and to evaluate their value forprognosis of the patients with gastric cancer.Methods (1) Immunohistochemical staining(PV-90002-step plus)was applied todetect the expression of XIAP Livin and Caspase-3in76cases of gastric cancer tissues、30cases of precancerous lesion tissues (including gastric polyp and gastriculcer) and30cases ofadjacent normal gastric mucosa.(2) The cell apoptosis of76cases of gastric cancer tissues wasdetected by TDT-mediated DNA in situ end labeling(TUNEL)method. The apoptotic index(AI)in XIAP Livin and Caspase-3positive and negative expressional groups were calculated andanalyze the relationship between survivin and Al.(3) The software package SPSS17.0wasused for statistical analysis.Results (1) The positive expression of XIAP in gastric cancer tissues was87.9%higher than30.0%in precancerous lesion tissues(P<0.05)and16.7%in adjacent normalgastric mucosa(P<0.05).(2) The positive expression of Livin in gastric cancer tissues was59.2%higher than20.0%in precancerous lesion tissues(P<0.05)and6.7%in adjacentnormal gastric mucosa(P<0.05).(3) The positive expression of Caspase-3in gastriccancer tissues was32.9%lower than66.7%in precancerous lesion tissues(P<0.05)and76.6%in adjacent normal gastric mucosa (P<0.05).(4) The expression of XIAP wasclosely correlated with tissue differentiation, lymph node metastasis (P<0.05),but not correlated with sex, age, tumor size,tumor depth and clinical stage of gastric cancer(P>0.05).(5) The expression of Livin was closely correlated with tissue differentiation,lymph node metastasis (P<0.05),but not correlated with sex, age,tumor size,tumor depthand clinical stage of gastric cancer (P>0.05).(6) The expression of Caspase-3was closelycorrelated with tissue differentiation (P<0.05),but not correlated with sex, age, tumorsize,tumor depth,lymph node metastasis and clinical stage of gastric cancer (P>0.05).(7)By Spearman analysis, XIAP expression was not associated with Livin proteinexpression in gastric cancer(γ=0.529, P>0.05).There was significant negative correlationbetween the expression of XIAP Livin and Caspase-3expression(γ=0.529, P<0.05).(8) Theapoptotic index(Al) was1.8%-3.5%and the overall average was(5.16士3.86)%. Averageapoptosis index of XIAP protein positive gastric cancer group was significantly lower than thatof XIAP protein negative group (P<0.05). Average apoptosis index of Livin protein positivegastric cancer group was significantly lower than that of Livin protein negative group (P<0.05).Average apoptosis index of caspase-3protein positive gastric cancer group was significantlyhigher than that of caspase-3protein negative group (P<0.05).(9) The total five-yearsurvival rate was46.8%in56cases of patients. Univariant analysis revealed that the survivalrate was correlated with expression of XIAP, Livin, caspase-3histological grade and TNMstage, as well as lymph node metastasis (P<0.05), but sex, age, tumor location, size, wereunrelated to prognosis (P>0.05); Multivariate analysis showed that it was lymph nodemetastasis and Livin were independent prognostic factor for patients with gastric cancer.Conclusions (1) The abnormal expression of XIAP Livin and Caspase-3protein ingastric cancer tissues may play a role in the occurrence and development of gastric cancer.(2)There was no correlation between the expression of XIAP and Livin in gastric cancer tissues. Itindicated that there could be a different mechanism in the cells apoptosis.(3) Lymph nodemetastasis and the expression level of Livin were independent prognostic factor for patientswith gastric cancer, but XIAP and Caspase-3were not.(4) Livin served as a new target for apoptosis-inducing therapy of gastric cancer.
