To Observe HRS Cells Of Hodgkin Lymphoma Combined With16Antibodies By AFM

Objective: Using AFM directly observed the HRS cells in combination withantibodies，the changes of numbers cell surface antibody particles reveal clues of theHRS cells source.Methods: With steam method of10%formalin fixed，17kinds of antibodiesobserved under the AFM scanning. Fresh lymph node tissues of patients with HLremoved surgically were printed.17kinds of antibody respectively were dropped onthe printed slides. According to conventional immunohistochemical methods, slideswere dried and observed by the application of AFM Tapping mode. The HRS incombination with antibodies after scanning, the surface membrane structures wereshowed clear images. Using AFM software analysis system for measurement andanalysis of the number of particles on the cell membrane surface, it is concluded andcounted.Results:1, A case was selected by the classical Hodgkin lymphoma，mixedcellular subtype.2, The particles of HRS cells combined with17kinds of antibodywere counted and the numbers were between3-46. Three particles: printed piecewithout antibody (negative control), CK, CD38; Five particles: CD3;6particles:CD20, CD30; Nine particles: CD5, CD10;13particles: CD35;14particles: CD15,CD163;16particles: the BCL-2;17particles: PAX-5;23particles: MUM-1;24particles: LCA;34particles: CD21;46particles: CD68.Conclusion: The cell surface morphology application of AFM scanning17kinds ofantibody combined with HRS cells were observed clearly. The antibody particlenumber can be divided into three categories: less particle (less than10), the middleparticle (11-20) and more particles (more than21). This results directly confirm thepolymorphism of the HRS cells combined with different antibodies.The HRS cells in combination with antibodies of B lymphocytes, T lymphocytes,dendritic cells and histiocytes and the different numbers of antibody particles promptHRS cells have a variety of lymphoid hematopoietic tissue antigen expression on the cell membrane. It is an obviously clue for the source research of HRS cell under themorphological basis.
