Hepatocellular carcinoma is one of the most common malignant tumorsworldwide. The occurrence of Hepatocellular carcinoma is associated with manyfactors,the majoy risk factor for HCC in China is Hepatitis B virus infection. Theoccurrence of HCC is a pathology process with many gene participation、 multiplesteps and stages. The vast majority of liver cancer patients have been infectedhepatitis B virus in China. In the process of occurrence and development of HCC,themechanism of HBV-mediated HCC is extremely complex and not clear. A lotof evidence support the notion that Hepatitis B virus X gene plays an important rolein the transformation of HCC.In recent years, research shows X gene is closely related to the transformationof HCC. X gene is the smallest open read frame of HBV DNA. HBx is a kind ofmulti-functional protein and involved in multiple steps of hepatocellular carcinomadevelopment. With the use of advanced molecular biology technology,plenty ofachievement has been obtained in the study of the role of HBx in HCC developmentincluding the following:the transactivation function、the activation signal transductionpathways、the influence of P53and the regulation of cell proliferation and apoptosis.Our country is a area of high Hepatitis B virus prevalence.Therefore,studying the relationship between HBx and HCC has an important theoretical andpractical significance for the prevention and treatment of hepatocellular carcinoma.Part I Expression of HBx protein and HBc protein in41HCCtissues and adjacent-tumor tissuesObjective To investigate the expression of HBx protein and HBc protein in41HCC tissues and adjacent-tumor tissuesMethods The expression of HBx protein and HBc protein in41HCC tissuesand adjacent-tumor tissues were detected by immunohistochemistry.Result There were33positive cases(80.4%)in HCC tissues and37positivecases(90.2%)in adjacent-tumor tissues without significant difference(P>0.05).The positive expression of HBx protein and HBc protein in41HCC tissues was80.4%(33/41)and29.2%(12/41).It was90.2%(37/41)and48.7%(20/41)inadjacent-tumor tissues. Difference was significant(P<0.05).Part II Relationship between the expression of HBx protein andHBVDNA in hepatocellular carcinoma tissueObjective To investigate the relationship between the expression of HBx proteinand HBVDNA in hepatocellular carcinoma tissueMethods The expression of HBx protein was detected by immunohistoche-mistry. And the expression levels of hepatitis B virus DNA were measured byquantitative PCR.Result The positive expression of HBx protein tissues was17(73.9%) andpositive cases of hepatitis B virus DNA was19(82.6%) in23HCC tissues.Therewas a positive correlation between HBx protein expression and the prevalence ofHBV DNA(r=0.455,P<0.05).Conclusion Combine the above two parts of the experimental results,there isrelationship between the expression of HBx protein and pathogenesis of HCC.Theexpression of HBx protein and the level of HBV DNA may play important role inthe occurrence of HCC.
